摘 要:为了探讨四棱豆肽组分的抗疲劳效果及对抗疲劳肽进行分离和鉴定,采用碱性蛋白酶对四棱豆蛋白进行酶解,经反相液相色谱分离后,得到Sample1、Sample2和Sample3 3个主要组分,分别以1.5 g/kg体重的剂量灌胃小鼠。结果表明:经游泳耐力实验,3个组分都可以延缓小鼠的游泳疲劳,但与四棱豆蛋白组相比,仅Sample3肽成分可以显著延缓小鼠的游泳疲劳,提高血浆中葡萄糖和游离脂肪酸的含量和肝糖原的水平。通过氨基酸组成分析,Sample3中富含抗氧化的疏水氨基酸(Leu、Ala、Ile、Pro、Val和Phe),可以有效延缓由自由基引发的疲劳。经过色谱分离,得到Sample3肽成分中一个优势的小肽S1,其氨基酸序列为Gln-Ser-Pro-ProGlu-Ile-Asn-Thr-Val,符合抗疲劳肽的特点。灌胃小鼠sample3组分和分子量小于1 000的米渣蛋白肽,疲劳前的游泳时间没有显著性差异,灌胃肽S1后的小鼠疲劳前的游泳时间显著增加。说明肽S1是一种潜在的天然抗疲劳物质。In order to explore the effect of a peptide with high anti-fatigue from Psophocarpus tetragonolobus (L.) DC., the peptide was separates and characterized after alkalase hydrolysis. Three main components, Samplel, Sample2 and Sample3 were separated by RP-HPLC. The swimming fatigue time could extend significantly compared with that in vehicle group in swimming endurance test for the mice orally administered three components a dose of 1.5 g/kg body weight respectively. Compared with the P. tetragonolobus protein group, only the Sample3 group could significantly prolong the swimming fatigue time, and increase the content of glucose and free fatty acids in plasma and the level of hepatic glycogen. By amino acid composition analysis, Sample3 contained abundant antioxidant amino acids, such as Leu, Ala, Ile, Pro, Val and Phe (hydrophobic amino acid), and could effectively delay the fatigue induced by free radicals. After chromatographic separation, a small peptides S1 with mino acid sequence Gln-Ser-Pro-Pro-Glu-Ile-Asn-Thr-Val was identified from Sample3, which was in line with the characteristics of anti-fatigue peptide. There were no significant differences in swimming fatigue time in Sample3 group and rice residue peptide group with molecular weight of less than 1 000. The swimming fatigue time of mice in peptide S1 group increased significantly. Peptide S1 could be used as a natural anti-fatigue substance.
