摘  要:目的探索3.0T MR活体示踪移植SPIO标记骨髓间充质干细胞(BMSCs)在大鼠脊髓损伤(SCI)模型中的优选序列。方法采用贴壁法培养大鼠BMSCs,使用超低微浓度的菲立磁-硫酸鱼精蛋白(Fe-Pro)标记BMSCs,普鲁士蓝及核固红染色显示细胞内铁。将20只脊髓损伤(SCI)模型的SD大鼠,于建模后第7天在损伤区两侧局部多点注射Fe-Pro标记的BMSCs,并于移植后第14天行3.0T MR FSET2WI、3DFIESTA、ESWAN序列成像,测量损伤区标记干细胞信号变化率及图像信噪比,并与脊髓组织切片普鲁士蓝染色对照。结果 20只SD大鼠BMSCs标记率为100%。脊髓普鲁士蓝染色显示,损伤区附近细胞质内大量细小蓝染颗粒。标记干细胞在3种MR序列图像上的信号变化率分别为(43.40±3.20)%、(77.80±3.59)%、(79.60±3.58)%,组间比较差异有统计学意义(P<0.05),FSET2WI序列的信号变化率均小于3DFIESTA及ESWAN序列的信号变化率,差异有统计学意义(P<0.05);3DFIESTA及ESWAN序列间信号变化率比较差异无统计学意义(P>0.05)。标记干细胞在3种MR序列图像信噪比分别为(20.95±1.91)、(45.70±9.40)、(44.50±8.29),组间比较差异有统计学意义(P<0.05),FSET2WI序列的信噪比均小于3DFIESTA及ESWAN序列的信噪比,差异有统计学意义(P>0.05);3DFIESTA及ESWAN序列间信噪比比较差异无统计学意义(P>0.05)。结论 3.0 T MR FSET2WI、3DFIESTA、ESWAN序列均可以检测到SPIO标记的BMSCs在大鼠脊髓内的MR信号,ESWAN序列效果最佳。Objective To explore a optimum 3.0 T MR sequence tor tracing the late of SPIO-labeled bone marrow mesenchymal stem cells(BMSCs) in vivo.Method BMSCs were isolated and expanded by means of differential adhesion method and then labeled with Fe-Pro complexes.Prussian and Nuclear Fast Red were performed for showing intracellular irons.Fe-Pro labeled BMSCs was locally injected into both sides of the damaged areas in multiple spots into the 20 spinal cord injury(SCI) model rats 7 days after the injury.3.0TMR FRFSET2 WI,3DFIESTA,ESWAN sequences were performed to gain MR imaging 14 days after transplantation,then signal-to-noise ratio(SNR) of spinal cord and signal rate of labeled cells as well as comparison with Prussian traced spinal cord tissue were obtained.Result In the 20 SD rats,BMSCs was 100%labeled.Prussian blue staining:Iron-containing intracytoplasmic vesicles can be observed clearly in cytoplasmic nearby injury with Prussian blue staining.The signal rate of labeled cells was(43.40 ± 3.20)%,(77.80 ± 3.59)%and(79.60 ± 3.58)%respectively on the three sequences where there were significant differences in group comparisons(P0.05).The signal changing rate of FSET2 WI sequence was below than that of 3DFIESTA and ESWAN sequences(P〈0.05),while there was no significant difference between the signal changing rate of 3DFIESTA and that of ESWAN sequences(P〈0.05).The SNR of labeled stem cells were(20.95 ± 1.91),(45.70 ± 9.40) and(44.50 ± 8.29) respectively on the three sequences where there were significant differences in group comparison(P〈0.05),in which the SNR of FSET2 WI sequence was lower than that of 3DFIESTA and ESWAN sequences(P〈0.05),while there was no significant difference between the SNR of 3DFIESTA and ESWAN sequences(P〈0.05).Conclusion The MR signal of SPIO-labeled BMSCs can be detected by 3.0T MR FRFSE T2 WI,3DFIESTA,and ESWAN sequences.Moreover,ESWAN sequences show best effect.
