摘  要:目的高效液相色谱法(HPLC法)测定阿图什无花果叶汁中伞形花内酯、佛手柑内酯和补骨脂素3种香豆素的含量。方法色谱柱Sunfire C_(18)ODS(250 mm×4.6 mm,5μm),流速1 mL、min,检测波长为220 nm,柱温30℃,流动相为乙腈-水,流动相配比为0~10 min,乙腈25%;10~12min,乙腈由25%梯度增至50%;12~20 min,乙腈50%;20~22 min,乙腈由50%梯度增至100%。结果伞形花内酯、佛手柑内酯和补骨脂素分别在0.001~0.019 mg、mL(R^2=0.999 9)、0.01~0.19 mg、mL(R^2=0.9997)和0.01~0.19 mg、mL(R^2=0.9999)范围内与峰面积呈良好线性关系,平均回收率分别为98.09%(RSD=0.85%)、102.07%(RSD=1.07%)和101.00%(RSD=0.71%)。阿图什无花果叶汁中伞形花内酯、佛手柑内酯和补骨脂素3种香豆素的含量分别为1.09、9.89、24.58 mg/g。结论HPLC法简便、准确、稳定,可用于测定无花果叶汁中伞形花内酯、佛手柑内酯和补骨脂素的含量。Objective To determine the contents of Umbelliferone,Bergapten and Psoralen in Ficuscarica Latex by HPLC method.Methods HPLC method conditions and procedure:Sunfire C_(18) ODS column(250 mm×4.6 mm,5 μm),with the flow rate of 1.0 mL/min,detectional wavelength at 220 nm under column temperature 30°C;acetonitrile-H_2O as the mobile phase,0~10 min,acetonitrile as 25%;afeter10~ 12 min,acetonitrile as going up to 25%~ 50%gradually;12~ 20 min,acetonitrile as 50%;20~22 min,acetonitrile as going up to 50%~100%gradually.Results There was a good linear relationship over the concentration range of(0.001~0.019) mg/ml,R^2 =0.999 9 for Umbelliferone,(0.01~0.19) mg/ml,R^2 =0.9997 for Bergapten and(0.01~0.19) mg/ml,R^2 =0.9999 for Psoralen.The average recovery rates of Umbelliferone,Bergapten and Psoralen were 98.09%(RSD = 0.85%),102.07%(RSD=1.07%) and101.00%(RSD=0.71%),respectively.The contents of Umbelliferone,Bergapten and Psoralen were1.09,9.89 and 24.58 mg/g.Conclusion The method is simple,accurate and stable,which could be applied to the determination of quality control.
