趋化型肿瘤细胞疫苗B16-MCP-1对小鼠恶性黑色素瘤免疫效果研究

摘　　要：目的构建小鼠恶性黑色素瘤动物模型,研究该疫苗在体内是否具有抗肿瘤活性,初步探讨其发生机制。方法将45只裸鼠随机分三组,每组15只:第一组为生理盐水空白对照组(Na Cl组),第二组为空载体p EGFP-N1转染细胞对照组(野生型B16疫苗组),第三组为趋化型肿瘤疫苗B16-MCP-1治疗组(趋化型B16-MCP-1疫苗组)。通过胸腺T淋巴细胞移植重建裸鼠部分免疫功能,观察荷瘤小鼠的一般情况、肿瘤大小、生存时间,分离并测定小鼠脾脏细胞中CTL的杀伤活性。结果趋化型肿瘤细胞疫苗B16-MCP-1组裸鼠生存状态明显好于空白对照组及野生型B16疫苗组(第一组和第二组裸鼠出现明显消瘦、目光呆滞、活动减弱等恶病质现象,第三组裸鼠生存状态明显好于前两组裸鼠);2周后第三组裸鼠肿瘤体积明显小于第一组及第二组(P<0.05);第三组裸鼠至35 d开始出现死亡,至观察结束(80 d)仍有4只裸鼠存活,与第一、二组比较生存时间明显延长(P<0.01);第三组淋巴细胞出现明显的特异性杀伤肿瘤细胞的活性,与第一组及第二组相比差异具有统计学意义(P<0.01)。结论该趋化型疫苗对裸鼠恶性黑色素瘤具有明显的免疫效果,能减轻荷瘤裸鼠的症状,延长裸鼠的生存期。ObjectiveThrough constructing the nude mouse malignant melanoma animal models to study whether the vaccine has theantineoplastic activityin vivo, and to explore its mechanism. MethodsPart of the immune function of the nude mouse was rebuild by transplanting thymic T cells of the same type of mouse. Observation was made on general condition of nude mice with melanoma, tumor size and survival time; CTL was from spleen cells of nude mice and its killing activity was determined. ResultsThe survival state of the group with B16-MCP-1 chemotactic tumor vaccine was significantly better than the control group and wild type B16 group and the tumor size was significantly smaller (P<0.05), survival time was significantly longer (P<0.01); the spleen cells had significantly specific killing activity in the B16-MCP-1 group (P<0.01).ConclusionThe chemotactic tumor vaccine has a significant immune effect on nude mice bearing melanoma, and can alleviate the symptoms of the tumor-bearing nude mice and prolong their survival time.
