摘  要:目的:探讨姜黄素对人乳腺癌细胞T47D增殖与凋亡的影响及机制。方法:不同浓度(20,40,80μmol/L)姜黄素作用于体外培养的人乳腺癌细胞T47D,采用MTT法分析细胞增殖抑制,流式细胞术分析细胞凋亡,RT-PCR检测Bcl-2mRNA表达。结果:姜黄素作用后,T47D细胞的生长抑制率明显上升,呈剂量和时间依赖性,细胞凋亡率增高,Bcl-2 mRNA表达水平降低。结论:姜黄素诱导T47D凋亡,抑制T47D生长,其机制可能与其下调Bcl-2mRNA表达有关。Objective:To investigate the effect of curcumin on the proliferation and apoptosis of T47D cells and its possible mechanism.Methods:Human breast cancer T47 Dcells were cultured and treated by curcumin in different concentrations(20,40 and 80μmol/L)in vitro.MTT assay was applied to determine the effect of curcumin on proliferation level of T47 Dcells,while apoptosis rate was assessed by flow cytometry.The mRNA level of Bcl-2 was measured by RT-PCR.Results:After curcumin treatment,MTT assay indicated that curcumin could inhibit the growth and proliferation of T47 Dcells in a time and concentration-dependent manner,and flow cytometry showed that apoptotic rate increased significantly.Furthermore,curcumin resulted in the obviously reduced expression of Bcl-2 mRNA in T47D cells.Conclusion:Curcumin can inhibit the growth of human breast cancer T47D cells and inducing apoptosis the mechanism may be related with the inhibition of Bcl-2mRNA expression.
