MicroRNA-146a基因多态性与慢性乙型肝炎患者抗病毒疗效的相关性

摘　　要：目的探讨micro RNA-146a(mi R-146a)基因单核苷酸多态性(SNP)和慢性乙型病毒性肝炎(CHB)患者干扰素-α(IFN-α)治疗应答反应的相关性。方法选择接受IFN-α抗病毒治疗的慢性乙型肝炎患者133例,其中应答组79例,无应答组54例。采集两组静脉血并提取DNA,用聚合酶链式反应-限制性内切酶长度多态性(PCR-RFLP)方法对mi R-146a行SNP分型。结果 mi R-146a CT基因型与TT基因型比较,差异有统计学意义(P=0.009);CC基因型与TT基因型比较,差异有统计学意义(P=0.029)。T等位基因与C等位基因频率比较差异有统计学意义(P=0.020)。结论 Mi R-146a基因多态性与慢性乙型肝炎患者抗病毒疗效相关,T等位基因或TT基因型与IFN治疗应答相关,而C等位基因或CC基因型与干扰素治疗应答无相关。[objertive] To investigate the relationship between the single nucleotide polymorphism （SNP） on miR-146a gene and the treatment effects of TFN-α. [methods] Totally 133 patients of chronic hepatitis B infection who received the treatment of IFN-α were enrolled in this study. The patients were divided into two groups： sustained virological response （n = 79） group and non virological response （NVR） or recurrence （n = 54） group. Blood samples were collected and chromosomal DNA was extracted. The miR-146a polymorphism was determined with the method of polymerase chain reaction restriction fragment length polymorphism （PCR- RFLP）. [Results] In our study, there was a statistical association between miR-146a polymorphism and the antiviral therapy efficacy in the hepatitis B patients. There was a statistical difference in the CT genotype and the TF genotype of miR-146a between the two groups （P= 0.009）. There was a significant difference in the CC genotype and Tr genotype between the two groups （P= 0.029）. There was a significant difference in the C allele and T allele between the two groups （P= 0.02）. [Conclusions] These findings suggested that the SNP in miR-146a is associated with the antiviral therapy efficacy of hepatitis B patients. And the TT genotype or T alleles is associated with the response to TFN-α therapy while the CC genotype or C allele is not related to the response to TFN-α therapy or recurrence.
