摘 要:分别使用3种复合诱变方法处理链霉菌Snea253-GL 8菌株,比较3种诱变方法的致死率和正突变率,测定突变株的遗传稳定性及代谢产物的杀线虫活性,并筛选稳定高产的突变株。结果表明:UV+EMS诱变的正突变率最高为38.4%,致死率78.9%,杀线虫活性为93.0%;UV+DES诱变的正突变率为33.7%,致死率为73.8%,杀线虫活性为92.7%;UV+LiCl诱变的正突变率为29.5%,致死率为82.0%,杀线虫活性为93.7%。5代稳定遗传结果表明,各代之间的杀线虫活性差异不显著,遗传稳定性好。Three compound mutation methods were used to deal with Streptomyces Snea253-GL 8 strain and the mortality rate and positive mutation rate of 3 mutation methods were compared.The genetic stability and nematicidal activity of mutant strains were determined to screen stable and high yield mutant strains.The results indicated that the positive mutation rate of UV + EMS treatment was 38.4%,the lethality rate was 78.9%,and the nematicidal activity was 93.0%;the positive mutation rate of UV + DES treatment was 33.7%,the lethality rate was 73.8%,and the nematicidal activity was 92.7%;the positive mutation rate of UV +LiCl treatment was29.5%,the lethality rate was 82.08%,and the nematicidal activity was 93.7%.The results of genetic stability of 5 generations showed that there were no significant differences of nematicidal activity between each generation,and the genetic stability was good.
