合成的鹅去氧胆酸衍生物HS-1200抑制体外肝星状细胞增殖及Ⅰ型胶原蛋白合成的研究

摘　　要：目的研究合成的鹅去氧胆酸衍生物HS-1200抑制体外肝星状细胞增殖及I型胶原蛋白合成的效果。方法用5ng/ml TGF-β1刺激HSC-T6细胞,然后在24h,48h时给予不同浓度梯度的HS-1200(60μM,40μM,20μM)。本实验使用MTT法测定细胞增殖,采用流式细胞检测细胞周期。免疫组织化学研究Ⅰ型胶原蛋白、cyclin-D1、cyclin-E的表达;采用DNA片段分析研究细胞凋亡。结果本研究发现HS-1200抑制TGF-β1诱导HSC的增殖且将细胞捕获在细胞周期的G_0/G_1期。结论 HS-1200抑制HSC-T6增殖及I型胶原蛋白合成,具有作为临床抗纤维化试剂的潜力。Objective To research the effect of HS-1200 on the proliferation and the secretion of type I collagen of hepatic stellate cell by Induction of TGF-β1.Methods HSC-T6 cells were treated with different concentrations of HS-1200（60μM,40μM,20μM）for 24 hours and 48 hours,5ng/ml TGF-β1is added for stimulating.The effect of HS-1200 on HSC-T6 proliferation is analyzed with MTT colorimetry and cell periodic location is examined by Flow Cytometry（FCM）;expressions of type I collagen and cyclin D1,cyclin E protein is examined by immunocytochemistry.Apoptosis of HSC-T6 is detected by DNA fragmentation.Results HS-1200 can remarkably inhibit the stimulation of TGF-β1to HSC-T6 proliferation and induce an increase of HSC-T6 in G0/G1 stage.Conclusion HS-1200 can remarkably inhibit the expression of type I collagen and induce apoptosis of HSC-T6 cells stimulated by TGF-β1.
