摘 要:目的:建立叠鞘石斛中总酚的含量测定方法,并对叠鞘石斛中的总酚含量进行测定。方法:考察磷钼酸显色法、普鲁士蓝显色法和福林酚法3种显色方法对叠鞘石斛中总酚定量分析的适用性,通过L_9(3~4)正交实验选择最佳的显色条件。结果:福林酚法适用于叠鞘石斛中总酚含量的测定;显色条件:检测波长760nm,福林酚试剂用量为1 mL,10%Na_2CO_3溶液用量为5 mL,反应时间为1 h,反应温度为30℃,线性范围为4~12μg·mL^(-1),平均加样回收率为98.14%,RSD为1.64%;总酚平均含量为6.75~7.85 mg/g。结论:该方法可用于叠鞘石斛中总酚含量的测定,且简单易行,稳定可靠,重现性好。Objective:To establish a UV method for determination of total hydroxybenzene for Dendrobinm denneanum.Methods:Phosphomolybdic acid colorimetric method,Prussian blue and FolinDenis are commonly used in the determination of total hydroxybenzene,we chose it as the best method which has a suitable maximum absorption.The conditions of the colorimetric method were investigated,then the best condition is based on orthogonal test L_9(3~4).Results:Folin-Denis is superior to the other two methods,and colorimetric conditions were as follows:detection wavelength 760 nm,the amount of FolinDenis 1 mL,the amount of 10%sodium carbonate solution 5 mL,temperature 30℃,reaction time lh.The total hydroxybenzene at 4~12μg·mL^-1,r=0.9995,the average recovery was 98.14%,RSD=1.64%.Conclusion:The method is accurate and simple with good reproducibility,and can control the quality of this preparation effectively.
