摘　　要：目的:建立叠鞘石斛中总酚的含量测定方法,并对叠鞘石斛中的总酚含量进行测定。方法:考察磷钼酸显色法、普鲁士蓝显色法和福林酚法3种显色方法对叠鞘石斛中总酚定量分析的适用性,通过L_9(3~4)正交实验选择最佳的显色条件。结果:福林酚法适用于叠鞘石斛中总酚含量的测定;显色条件:检测波长760nm,福林酚试剂用量为1 mL,10%Na_2CO_3溶液用量为5 mL,反应时间为1 h,反应温度为30℃,线性范围为4~12μg·mL^(-1),平均加样回收率为98.14%,RSD为1.64%;总酚平均含量为6.75~7.85 mg/g。结论:该方法可用于叠鞘石斛中总酚含量的测定,且简单易行,稳定可靠,重现性好。Objective：To establish a UV method for determination of total hydroxybenzene for Dendrobinm denneanum.Methods：Phosphomolybdic acid colorimetric method,Prussian blue and FolinDenis are commonly used in the determination of total hydroxybenzene,we chose it as the best method which has a suitable maximum absorption.The conditions of the colorimetric method were investigated,then the best condition is based on orthogonal test L_9（3~4）.Results：Folin-Denis is superior to the other two methods,and colorimetric conditions were as follows：detection wavelength 760 nm,the amount of FolinDenis 1 mL,the amount of 10%sodium carbonate solution 5 mL,temperature 30℃,reaction time lh.The total hydroxybenzene at 4~12μg·mL^-1,r=0.9995,the average recovery was 98.14%,RSD=1.64%.Conclusion：The method is accurate and simple with good reproducibility,and can control the quality of this preparation effectively.
