摘  要:目的:研究卵巢癌细胞株SKOV3中ERβ表达对细胞增殖凋亡的影响。方法:MTT法检测卵巢癌细胞暴露于17β-E2不同浓度及不同时间的细胞增殖率。用特异性雌激素受体激动剂DPN联合17β-E2或单独使用DPN处理细胞,RT-PCR法检测ERβmRNA、ERK1/2 mRNA及caspase-3 mRNA表达,蛋白免疫印记法检测ERβ和p-ERK1/2蛋白表达。结果:10-6mol/L 17β-E2作用60min时,细胞的增殖活性最强(5.7±0.23),与对照组相比(3.5±0.45),差异有统计学意义(P<0.05)。ERβ在卵巢癌中低表达。随着DPN作用时间延长及浓度增加,ERβmRNA、caspase-3 mRNA表达逐渐增加(P<0.05),ERK2 mRNA表达逐渐降低,与对照组比较差异有统计学意义(P<0.05)。随着DPN作用时间延长及浓度的增加,ERβ蛋白表达逐渐增加,p-ERK1/2蛋白表达逐渐降低,与对照组比较差异有统计学意义(P<0.05)。结论:ERβ在卵巢癌中低表达,上调ERβ表达能抑制卵巢癌细胞的增殖,同时促进细胞的凋亡。Objective:To investigate the expression of estrogen receptor beta ( ER) ac-tivation on the proliferation and apoptosis in ovarian carcinoma cell. Methods:When the cells were exposed to 17β-E2 in different concentrations and for different time,the proliferation was assayed by MTT. After the cell was pretreated with DPN which was specific estrogen agonists and 17β-E2 or with DPN lonely,the expression of ERβ,caspase-3,ERK1,ERK2 mRNA were detected by RT-PCR,the protein expression of ERβ,p-ERK1/2 were detected by Western blot. Results:The cell proliferative rate was significantly higher in the cells(5. 7±0. 23) stimulated with 17β-E2 at 1"10-6mol/L for 60 min than that in the control ones(3. 5±0. 45)(P〈0. 05). The expression of ERβ decreased in the ovarian carcinoma. After the cells were treated with DPN,along with increasing of time and concentration, the expressions of ERβ and caspase-3 mRNA were increasingly higher than the control,while the expression of ERK1 mRNA,ERK2 mRNA were increasingly lower than that of control ( P〈0 . 05 ) . With increasing of time and con-centration,the protein expression of ERβ was increasingly higher than the control(P〈0. 05). While the protein expression of p-ERK1/2 was increasingly lower than the control(P〈0. 05). Conclusion:The expression of ERβ was decreased in ovarian carcinoma cells. When the ex-pression of ERβ was up-regulated, the proliferation was inhibited and the apoptosis was in-creased in ovarian carcinoma cell.
