摘 要:利用自行筛选的芽孢杆菌C2进行不同溶解氧环境的丁醇发酵试验,并对不同溶氧条件下微生物菌株胞内关键酶活性进行测定,以揭示溶氧对丁醇发酵的影响机制。结果表明:在兼性厌氧条件下丁醇和总溶剂的产量分别为9.50和13.02 g/L,比严格厌氧条件下丁醇(8.52 g/L)和总溶剂(11.74 g/L)产量分别提高了11.5%和9.01%。酶活性分析发现发酵对数期兼性厌氧条件下乙醇脱氢酶、丁醇脱氢酶、丁醛脱氢酶、乙酰乙酰辅酶A转移酶、乙酸激酶、丁酸激酶活性低于严格厌氧条件下其酶活性。然而发酵24 h之后,兼性厌氧条件下发酵的酶活性逐步升高,超越严格厌氧条件酶活性且持续增长至36 h后开始缓慢降低。芽孢杆菌C2的研究为生物丁醇的兼性厌氧发酵提供了宝贵的菌种资源和技术支撑。Strictly anaerobic environment is an important factor to impose restriction for butanol industrialization. Under different dissolved oxygen conditions, this study was carried out with the Bacillus sp. C2 screened in our lab. The intracellular key enzyme activities of strains grown under different conditions were detected in order to reveal the influence of dissolved oxygen on mechanism of butanol fermentation. The experiment results indicated that in butanol concentration (9.50 g/L) and total solvents concentration (13.02 g/L) under faeultative anaerobic conditions were respectively increased l l. 5% and 9.01% compared with butanol concentration (8.52 g/L) and total solvents con- centration (ll. 74 g/L) under strictly anaerobic conditions. The specific activity of alcohol dehydrogenase, butanol dehydrogenase, butylaldehyde dehydrogenase, acetylacetyl-CoA transferase, kinase acetate, butyrate kinase under facultative anaerobic conditions were lower than strictly anaerobic conditions at the lag phase of fermentation. Howev- er, with the extension of fermentation time, the enzyme activity under facultative anaerobic conditions was increased and beyond the strictly anaerobic conditions from 24 h and began to decrease slowly after 36 h of fermentation. This research provided a valuable strain and would be beneficial for bio-butanol industrialization.
