摘  要:目的观察高盐诱导的高血压大鼠炎症因子和内皮损伤因子的表达水平并探讨其与高血压的相关性。方法选取8周龄Wistar大鼠80只,随机分为对照组(30只)和模型组(50只),分别给予正常饮水和2%(质量分数)Na Cl高盐饮水,喂养21天,测量大鼠尾压以及血流动力学指标;采用酶联免疫吸附试验测定大鼠血浆高敏C反应蛋白(hs-CRP)、单核细胞趋化蛋白1(MCP-1)、肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)、血管性假血友病因子(v WF)、6-酮前列环素(6-K-PG)、内皮型一氧化氮合酶(e NOS)、一氧化氮(NO)、内皮素l(ET-1)的含量。结果高盐饮食21天后,模型组大鼠血压显著升高,盐敏感高血压动物模型构建成功。模型组大鼠左心室质量指数(LVMI)、左心室舒张期末压(LVEDP)与对照组相比有明显升高,而左心室压最大上升/下降速率(±dp/dtmax)降低,两组差异有统计学意义(P<0.05);而实验后左心室收缩压(LVSP)比较两组差异无统计学意义(P>0.05)。与对照组相比,模型组血浆中炎症因子hs-CRP、MCP-1、TNF-α及IL-6水平显著升高(P<0.05)。相关性分析显示,hsCRP、MCP-1、TNF-α及IL-6与血压均呈正相关(r分别为0.731、0.583、0.858、0.836,P<0.05)。v WF和ET-1浓度也显著增加,其水平与血压正相关(r=0.680,0.739,P<0.05)。血管舒张因子NO、6-K-PG浓度显著降低,e NOS水平也显著降低,且与血压呈负相关(r分别为-0.710、-0.658、-0.721,P<0.05)。结论盐敏感高血压大鼠存在血管炎症反应和内皮功能损伤。Aim To investigate the role of inflammatory factor and endothelial damage factor in high-salt induced hypertensive rat model. Methods Wistar rats with 8-week old were randomly divided into control group( fed with tap water,n = 30) and model group( fed with high-salt water containing 2% Na Cl,n = 50). After fed for 21 days,the blood pressure and hemodynamic parameters were determined. The high-sensitivity C-reactive protein( hs-CRP),monocyte chemotactic protein-1( MCP-1),tumor necrosis factor-α( TNF-α),interleukin-6( IL-6),von Willebrand factor( v WF),6-keto-prostaglandin( 6-K-PG),endothelial nitric oxide synthase( e NOS),nitric oxide( NO) and endothelin-1( ET-1)were detected by the ELISA method. Results After 21 days of high salt dietary,the blood pressure in model group was significantly higher than blank control group. The left ventricular mass index( LVMI) and left ventricular end diastolic pressure( LVEDP) in model group were significantly higher than control group,and the maximal rate of rise and fall of the ventricular pressure( ± dp / dtmax) was lower than control group after experiment( P〈0. 05). Compared with control group,the hs-CRP,MCP-1,TNF-α and IL-6 in blood plasma were significantly increased in model group,and the levels of the hs-CRP,MCP-1,TNF-α and IL-6 were found to be positively correlated with blood pressure( r = 0. 731,0. 583,0. 858,0. 836,P〈0. 05). The v WF and ET-1 were increased and positively correlated with blood pressure too( r =0. 680,0. 739,P〈0. 05). The NO,6-K-PG and e NOS were reduced in model group,and the levels of NO,6-K-PG and e NOS were negatively correlated with blood pressure( r =- 0. 710,- 0. 658,- 0. 721,P〈0. 05). Conclusion After fed with high-salt water for 3 weeks,the blood pressure of model group elevated. There are inflammatory reaction and damage of endothelium in model group.
