摘  要:目的探讨梗阻性黄疸模型小鼠自然杀伤(natural killer,NK)细胞的水平变化及意义。方法 80只小鼠随机分为手术组和假手术组各40只,手术组行胆总管结扎术制备梗阻性黄疸小鼠模型,假手术组仅游离胆管。分别于第7天和第14天取小鼠血清及肝脏组织,检测2组血清谷丙转氨酶、谷草转氨酶、总胆红素、直接胆红素和间接胆红素水平;对肝脏组织行组织病理检查HE染色、Masson三染色,采用免疫组织化学法检测α-平滑肌肌动蛋白表达情况,采用实时荧光定量PCR检测肝脏NK细胞表面特异性标志NK1.1、肿瘤坏死因子(tumor necrosis factor,TNF)-α和TNF相关凋亡诱导配体(TNF-related apoptosis inducing ligand,TRAIL)mRNA表达情况。结果手术组第7、14天血清谷丙转氨酶、谷草转氨酶、总胆红素、直接胆红素和间接胆红素水平较假手术组明显升高(P<0.01),第14天各指标水平高于第7天(P<0.05);手术组第7天NK1.1、TRAIL、TNF-αmRNA相对表达量(36.04±4.42、1.96±0.29、203.23±20.90)明显高于假手术组(1.00±0.00、1.00±0.00、1.00±0.00)(P<0.01),第14天(168.69±12.90、4.95±0.43、530.44±57.66)明显高于第7天和假手术组(1.01±0.03、1.01±0.02、0.99±0.02)(P<0.01),假手术组第7天NK1.1、TRAIL、TNF-αmRNA相对表达量与第14天比较差异无统计学意义(P>0.05)。结论梗阻性黄疸模型小鼠肝脏NK细胞增多,与肝损伤严重程度明显相关,NK细胞可能通过TRAIL依赖途径参与梗阻性黄疸的肝损伤过程。Objective To investigate the change and its significance of hepatic natural killer (NK) cells in mice models with obstructive jaundice. Methods Eighty mice were randomly divided into operation group and sham-operation group, with 40 mice in each group. Operation group underwent common bile duct ligation to establish obstructive jaundice models, and sham-operation group only underwent separation of common bile duct. The levels of serum glutamate pyruvate transaminase (GPT), glutamic-oxaloacetic transaminase (GOT), total bilirubin (TBil), direct bilirubin (DBil) and indirect bilirubin (IBil) were detected in serum and liver tissue by day 7 and 14 after operation. The liver tissue received HE stain and Masson stain. The level of alpha-smooth muscle actin was detected by histopathologic method, and hepatic NK 1. 1 mRNA, tumor necrosis factor-alpha (TNF-a) mRNA and TNF-related apoptosis inducing ligand (TRAIL) mRNA were determined by real time fluorescence PCR. Results The levels of serum GPT, GOT, TBil, DBil and IBil were significantly higher in operation group than those in sham-operation group by day 7 and 14 after operation (P〈0.01), and were significantly higher by day 14 after operation than those by day 7 after operation (P〈0.05). The relative expression levels of NK1.1, TRAIL and TNF-a mRNA were the highest by day 14 after operation in operation group (168.69±12.90, 4.95±0.43, 530.44 ± 57.66), followed by those by day 7 after operation in operation group (36.04±4.42, 1.96±0.29, 203.23± 20.90), and by day 7 and 14 after operation in sham-operation group (1.00± 0.00, 1. 00±0. 00, 1.00±0.00; 1.01±0. 003, 1.01±0.02, 0.99±0.02) (P〈0.01), and there were no significant differences between day 7 after operation and by day 14 after operation in sham-operation group (P〉0.05). Conclusion Obstructive jaundice is commonly complicated with increased hepatic NK cells and it is obviously correlated with the severity of liver injury. NK cells might
