摘  要:目的建立标准的碱烧伤导致的角膜缘干细胞缺乏模型,并对模型进行评价,为研究临床治疗提供动物模型。方法实验研究。于2013年10月至2014年6月在山东中医药大学将18只新西兰大白兔随机分为A组:1mol/L氢氧化钠溶液浸泡的环形滤纸角膜缘贴附烧伤60s+50μl 1mol/L氢氧化钠溶液滴人结膜囊30s;B组:1mol/L氢氧化钠溶液浸泡的环形滤纸角膜缘贴附烧伤30s+50μl1mol/L氢氧化钠溶液滴人结膜囊30s,C组:1moUL氢氧化钠溶液浸泡的环形滤纸角膜缘贴附烧伤30S共3组。造模后观察各组新生血管长入角膜及睑球粘连情况,并行活体共聚焦显微镜、印迹细胞学、泪液分泌试验等检查。结果A组模型中6“只”眼均出现角膜穿孔、眼球萎缩。B组6“只”眼烧伤后4周时角膜新生血管长人,睑球粘连均为中度,而C组6“只”眼则无睑球粘连出现。B、C组12“只”眼:角膜缘新生血管在烧伤后1周开始长人,4周时几乎长满角膜;4周时,共焦显微镜检查示角膜缘典型的栅栏结构消失,角膜结膜化,出现杯状细胞,泪液分泌实验与对照眼差异有统计学意义(P〈0.05),B、C组之间差异无统计学意义(P〉0.05)。结论环形滤纸片浸润氢氧化钠溶液烧伤角膜缘组织,同时结膜囊滴人氢氧化钠溶液可以成功构建眼部碱烧伤导致角膜缘干细胞缺乏的动物模型。Objective To establish and evaluate an ocular alkali bum animal model of corneal limbal stem cell deficiency. Methods Eighteen New Zealand white rabbits were randomly divided into 3 groups: filter paper soaked with lmol/L NaOH solution affixed to the limbus for 60s, at the same time drop 50μl 1mol/L NaOH solution in the conjunctiva sac for 30s in group A; filter paper soaked with lmol/L NaOH solution affixed to the limbus for 30s, at the same time drop 50μ1 1mol/ L NaOH solution in the conjunctiva sac for 30s in group B; filter paper soaked with 1mol/L NaOH solution affixed to the limbus for 30s in group C. Model evaluation: new vessels into cornea and symblepharon was observed, in vivo confocal microscopy examination, impression cytology observation, Schirmer test. Results The all 6 eyes in group A were corneal perforated and ultimately atrophied. At week 4, 6 eyes in group B occurred corneal neovascularization and moderate symblepharon, 6 eyes in group C just occurred corneal neovascularization. Twelve eyes in group B, C, new vessels into cornea occurred at lweek, 4 weeks later cornea full with new vessels, at the sametime, in vivo laser confocal microscope examination showed limbal palisades of Vogt was disappeared, find goblet cells in the corneal epithelium, Schirmer Ⅰ test results were statistical different significantly between model and normal eyes. Conclusions Filter paper soaked with 1mol/L NaOH solution affixed to the limbus for 30s, at the same time drop 50μl 1mol/L NaOH solution in the conjunctiva sac for 30s can successfully established the rabbit ocular alkali bum model of corneal limbal stem cell deficiency.
