摘 要:目的:探讨定经汤调节卵巢早衰的作用机制。方法:SD大鼠按随机数字表法分为正常组、模型组、定经汤干预组,分别检测其与卵巢早衰相关的生物分子,如β-EP、IL-1、NOS、NO、Gn RH、CRH、FSH、LH、E2、P、ACTH和CORT;建立基于支持向量回归机的生物分子网络模型,探究定经汤干预下各生物分子之间相互调控关系。结果:卵巢早衰大鼠灌服定经汤后,垂体-卵巢轴及垂体-肾上腺轴靶腺层生物分子指标值上升,垂体-卵巢轴及肾上腺轴的变化反馈到下丘脑,使得IL-1、NOS和NO指标值上升;通过IL-1的枢纽作用,使得β-EP、Gn RH和CRH上升;最后下丘脑生物分子的上升又使得垂体-卵巢轴及肾上腺轴的靶腺层生物分子进一步上升,垂体-卵巢轴垂体层生物分子稍有下降。结论:定经汤干预下卵巢早衰相关生物分子网络趋于平衡,定经汤调节卵巢早衰过程中起到关键作用的生物分子为IL-1。Objective: Exploring the mechanisms of the premature ovarian failure( POF) regulation by Dingjing Decoction. Methods: 90 SD rats were randomly divided into three groups: normal groups,model groups and Dingjing Decoction groups; Biomolecules which associated with premature ovarian failure: β-EP,IL-1,NOS,NO,Gn RH,CRH,FSH,LH,E2,P,ACTH and CORT were detected. Regulation relations of biomolecules associated with POF regulation by Dingjing Decoction were explored based on support vector regression machine. Results: The values of biomolecules in pituitary-ovarian axis and target gland layer of pituitary-adrenal axis increased significantly in curing with Dingjing Decoction; then these changes feed back to the hypothalamus,leading the increase of the values of IL-1,NOS and NO in hypothalamus; then the values of β-EP、Gn RH and CRH increased in hypothalamus through the pivotal function of IL-1;The values of biomolecules in target gland layer of pituitary-adrenal axis and pituitary-ovarian axis increased significantly,the values of biomolecules in the pituitary layer of hypothalamic-pituitary-ovarian axis decreased slightly,when the values of biomolecules in hypothalamus increased. Conclusions: The regulation relations of biomolecules network associated with to be balance. IL-1 play a pivotal role in the biomolecules network associated with POF regulation by Dingjing Decoction.
