摘 要:以杏鲍菇深加工残渣为原料,利用单因素试验和Box-Behnken响应面法优化膳食纤维提取工艺;采用高效液相色谱-蒸发光散射法(HPLC-ELSD)分析膳食纤维的单糖组成。结果表明,酶解法提取膳食纤维的最佳工艺条件:反应温度40℃、p H7.0、中性蛋白酶添加量0.93%、液料比9.6:1、酶解时间4 h。在该优化条件下,膳食纤维得率32.15%,蛋白去除率86.53%。杏鲍菇深加工残渣中总膳食纤维含量为67.52%,主要以不溶性膳食纤维为主,其膳食纤维单糖主要由葡萄糖、甘露糖和阿拉伯糖组成,摩尔比为16.87:1.80:0.56。Single factor and response surface methodology was used to optimize the extraction of dietary fiber from residue of Pleurotus eryngii by enzyme hydrolyze method.The monosaccharide composition of dietary fiber from residue of Pleurotus eryngii was analyzed by high performance liquid chromatographyevaporative light scattering detection(HPLC-ELSD).The results showed that the optimal extraction conditions were 0.93%(m/m) enzyme content and the ratio of water to material 9.6:1 under the p H value 7.0 and the temperature 40 ℃ for an extraction time of 4 h.The yield of dietary fiber was 32.15% and the removal efficiency of protein was 86.53% under the optimal extraction condition.The proximate composition of dietary fiber showed that the content of total dietary fiber was 67.52% and the total dietary fiber was mainly made up of insoluble dietary fiber.The monosaccharide of dietary fiber consisted of glucose,mannose and arabinose,with the molar ratio of 16.87:1.80:0.56.
