摘  要:目的观察青蒿琥酯处理前后人肺腺癌PC9细胞增殖率的变化、对Axl与下游信号分子表达水平的变化及凋亡相关蛋白的影响。方法用不同终浓度的青蒿琥酯分别处理PC9细胞,24、48、72 h后通过cck-8检测细胞的增殖率。经25μmol/L及100μmol/L青蒿琥酯分别处理PC9细胞48 h后用qRT-PCR检测Axl、Akt mRNA的表达水平,用Western blot检测Axl、Akt及凋亡相关蛋白的表达水平。结果 1青蒿琥酯以剂量和时间依赖性地抑制PC9细胞的增殖,且凋亡相关蛋白caspase-3、PARP表达上调。2用药48 h后可以下调p-Axl、p-Akt的表达。结论青蒿琥酯能抑制肺腺癌细胞的增殖,导致凋亡相关蛋白表达上调,且能下调Axl及下游信号分子Akt mRNA和蛋白的表达。Objective To observe the changes of cell proliferation rate,apoptosis and Axl expression level before and after the treatment of Artesunate in human lung adenocarcinoma PC9 cells.Methods Different concentrations of Artesunate were used to treat PC9 cells.Cell proliferation was analyzed by the cck-8 assay after 24,48 and 72h.The final concentration of Artesunate treatment PC9 cells were 25,100 μmol/l,respectively.The mRNA expression levels of Axl and Akt were determined by qRT-PCR;The protein expression levels of Axl、Akt and apoptosis-related proteins by Western blot after 48 h.Results(1)Artesunate could inhibit PC9 cell proliferation dependent on the dose and time.The expression of caspase-3 and PARP increased after 48 h.(2)The mRNA and protein expression of p-Axl and p-Akt were reduced after 48 h.Conclusion Artesunate could inhibit the proliferation of lung adenocarcinoma cancer cells,up-regulate apoptosis-related proteins,and down-regulate the mRNA and protein expression of Axl and Akt,Which are downstream signaling molecules.
