摘  要:采用愈创木酚-PDA平板显色、Azure-B蓝色平板脱色初筛,摇瓶发酵复筛,从腐木中筛选到木质素酶高活性菌株Lys3002.经ITS测序分析和形态观察,Lys3002鉴定为毛栓菌(Trameteshirsuta).采用液体摇瓶优化发酵条件,结果表明:野生型菌株Lys3002的木质素酶系中漆酶的最适发酵条件是:麸皮20g/L,豆饼粉6g/L,培养基初始pH6.0,MgSO4·7H2O0.5g/L,CuSO4·5H2O0.05g/L,MnSO40.05g/L,培养温度30℃,发酵周期5d,漆酶活力高达597.82U/L,具有深入研究价值和应用潜力.A strain Lys3002 with high activity of ligninase was screened from a rotten wood by methods of guaiacol-PDA plate colorization and Azure-B blue plate decolorization and confirmed by fermentation in shake flask.Strain Lys3002 was identified as Trametes hirsuta by ITS sequencing analysis and morphological observation. Futher studies declared,by shake flask,the optimium fermentation conditions were as follows: wheat bran 20 g / L,soybean meal 6 g / L,initial medium p H 6. 0,Mg SO4·7H2O 0. 5 g / L,Cu SO4·5H2O 0. 05 g / L,Mn SO40. 05 g / L at 30 ℃for 5 days. The laccase activity reached 597. 82 U / L,which had application prospect and deserved further research.
