摘 要:从菌核分化入手,研究外源信号分子对汤姆青霉PT95菌株菌核分化的影响。使用MEA和PDA这2种不同的培养基培养汤姆青霉PT95菌株,并在2种不同的培养基上分别添加0,0.02,0.04,0.06 mmol/m L的c AMP;通过记录菌核成熟的时间,测量菌落直径、菌核生物量和类胡萝卜素含量,以分析菌株生长发育情况。结果表明,在加入的cAMP浓度达到0.02 mmol/m L时菌核的生长发育状态最佳;之后随着cAMP浓度的增加,其生长状况逐渐下降;当cAMP浓度达到0.06 mmol/m L时,其生长开始受到抑制,表明外界适当供给信号分子c AMP可促进汤姆青霉PT95菌株的生长,但cAMP浓度过高则会抑制汤姆青霉菌株的生长发育。The project was planned to start from sclerotia differentiation, effect of exogenous signal molecule on sclerotial differentiation of Penicillium thomii PT95 was studied. Using MEA and PDA two different culture medium of Penicillium thomii PT95,and 0, 0.02, 0.04, 0.06 mmol/m L of cAMP separately on two different media, by recording the time to sclerotia mature, measuring the colony diameter, sclerotia biomass and carotenoid content, to analyze the growth and development of the strains. The results showed that when the cAMP concentration reached 0.02 mmol/m L, the growth state of sclerotium was the best; after increases cAMP concentrations growth status decreased; when the cAMP concentration reached 0.06 mmol/m L, the growth began to be suppressed. That indicated that under the low concentration of cAMP, Penicillium thomii PT95 strain's growth was promoted. But if the supply of cAMP concentration was too high, sclerotia biomass and carotenoid content of PT95 strain would be lower than normal, the growth was inhibited.
