MTDH和MMP-9在喉鳞状细胞癌组织中的表达及意义

摘　　要：目的:观察喉鳞状细胞癌组织中异黏蛋白(MTDH)、基质金属蛋白酶-9(MMP-9)的表达变化,并探讨其意义。方法:采用免疫组织化学法检测54例喉鳞状细胞癌组织(观察组)、30例癌旁正常组织(对照组)中MTDH、MMP-9蛋白的表达,分析其与喉鳞状细胞癌临床病理参数的关系及二者的相关性。结果:观察组及对照组MTDH的阳性表达率分别为64.8%(35/54)、6.7%(2/30),差异有统计学意义(P<0.01);MMP-9的阳性表达率分别为70.4%(38/54)、13.3%(4/30),差异有统计学意义(P<0.01)。MTDH的阳性表达与喉鳞状细胞癌的分化程度、淋巴结转移及TNM分期有关(P<0.05),MMP-9的阳性表达与喉鳞状细胞癌淋巴结转移及TNM分期有关(P<0.05)。喉鳞状细胞癌组织中MTDH与MMP-9的表达呈正相关(r=0.371,P<0.01)。结论:MTDH和MMP-9的过度表达与喉鳞状细胞癌的发生、恶性发展及转移密切相关,联合检测2种蛋白的表达对喉鳞状细胞癌的早期诊断及预后判断具有一定的临床参考价值。Objective：To observe the expression changes of metadherin（MTDH）and matrix metalloproteinase-9（MMP-9）in the laryngeal squamous cell carcinoma tissues and to investigate the significance.Method：The expression of MTDH and MMP-9in 54 cases of laryngeal squamous cell carcinoma tissues（observation group）and30cases of para-carcinoma tissues（control group）was examined by immunohistochemical method,the correlation between them and their correlations with the clinicopathological parameters were analyzed.Result：The positive expression rates of MTDH in the observation group and control group were 64.8%（35/54）and 6.7%（2/30）,respectively;the positive expression rates of MMP-9in the observation group and control group were 70.4%（38/54）and 13.3%（4/30）,respectively;and there was a statistically significant difference between two groups（all P0.01）.In the laryngeal squamous cell carcinoma tissues,the expression of MTDH protein was related with degree of differentiation,lymph-node metastasis and TNM stage（all P0.05）;and the expression of MMP-9protein was related lymph-node metastasis and TNM stage（all P0.05）.The expression of MTDH was positively correlated with MMP-9in the laryngeal squamous cell carcinoma tissues（r=0.371,P0.01）.Conclusion：The high expression of MTDH and MMP-9was closely related to the occurrence,development and metastasis of laryngeal squamous cell carcinoma,joint detection of the two proteins was valuable for early diagnosis and prognosis of laryngeal squamous cell carcinoma.
