摘  要:目的优化鱼体内喹乙醇及代谢物的检测技术,探明兽药抗菌剂喹乙醇的生物富集特性。方法选择斑马鱼作为受试生物,通过检出效率试验确定了斑马鱼体内喹乙醇及代谢物MQCA的检测方法;采用半静态试验测定了2个暴露水平下喹乙醇的生物富集特性。结果乙腈超声提取.柱层析净化.高效液相色谱/质谱联用测定法(HPLC/MS)可同步检出斑马鱼体内喹乙醇及代谢标志物MQCA的含量;斑马鱼体内喹乙醇在的最大富集量分别出现在第4天(暴露浓度0.25mg/L)和第6天(暴露浓度1mg/L),生物富集系数为0.18~0.30,实验期内代谢物MQCA的最大残留浓度为0.013~0.016mg/kg,出现在第2天(喹乙醇暴露浓度1mg/L)和第4天(喹乙醇暴露浓度0.25mg/L)。结论本文建立的喹乙醇及其代谢物的检测技术准确可靠,单一水溶液暴露方式下斑马鱼对喹乙醇的富集性较低,本研究为该类药物的慢性毒性评价提供了基础数据支撑。Objective To optimize the determination techniques of olaquindox and its metabolite, and investigate the bioaccumulation characteristics of olaquindox. Methods The Brachydanio Rerio was selected as subject biological, and determination efficiency tests were conducted to ascertain the determinating methods of olaquindox and its MQCA metabolite. Moreover, semi static experiments in two olaquindox exposure levels were carried out to get the bioaccumulation characteristics. Results Techniques of ultrasonic extracting with acetonitrile and column purifying with purify columns filled with anhydrous zinc sulfate, and neutral alumina, and also anhydrous sodium, and HPLC/MS determinating was most efficient determination technics in determinating olaquindox and MQCA metabolic markers in Brachydanio rerio at the same time. In the experiments period of 8 days, the highest concentration of olaquindox in Brachydanio rerio appeard in the fourth day or the sixth day separately according to the 0.25mg/L or lmg/L exposure concentration, and the bioaccumulation factors of olaquindox in Brachydanio rerio were 0.18-0.30. At the same time, the maximum residue concentration of MQCA metabolite in fish was 0.013-0.016mg/kg, and happened in the second day and fourth day respectively according to the olaquindox exposure concentration of 0.25 or lmg/L. Conclusion These results indicated that the determination techniques of olaquindox and MQCA in Brachydanio rerio was practicable and credibility, and the bioaccumulation characteristics of olaquindox in Brachydanio rerio was relative lower in the case of single solution exposure, all above supplied the fundamental data for chronic toxicity evaluation of the drug on aquatic organism.
