摘 要:目的探讨基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)在多种肾小球疾病中的表达及意义。方法采用免疫组化Polink-2增强法检测MMP-9在增生性肾小球病变、基膜性肾小球病变和肾小球足细胞病变中的表达,分析表达位置及其在各种病变中的意义。结果 MMP-9在增生的系膜细胞、内皮细胞及新月体形成细胞胞质中呈强阳性,而系膜基质增生和纤维细胞性新月体形成时仅见少量弱阳性;MMP-9在基膜增厚病变为主的膜性肾病和Alport综合征肾小球足细胞胞质内呈强阳性;在薄基膜肾病肾小球中呈阴性;MMP-9在足细胞病肾小球中未见明显表达。结论 MMP-9参与炎症增生和肾小球基膜重建,辅助诊断肾小球基膜性病变。Purpose To investigate the expression of matrix metalloproteinase-9 ( MMP-9 ) in different renal diseases and its role on differential diagnosis. Methods The expression of MMP-9 in glomerular proliferative lesions, glomerular basement membrance lesions, and podocyte diseases were detected using immunohistochemistry method. Results The strong positive expression were found in cellular proliferative lesions, but weak positive expression in stromal proliferative lesions. The strong positive expression in podocyte were found in membranous nephropathy and Alport syndrome, but negative expression in thin basement membrane nephropathy. In podocyte diseases, the expression of MMP-9 is negative. Conclusion MMP-9 participate in inflammation and remodel of glomerular basement membrane, and expression of MMP-9 may helpful for the diagnosis of glomerular membranous lesions.
