摘 要:目的探讨葡萄胎病理相关新基因F10与绒癌细胞系JAR裸鼠皮下成瘤的关系。方法通过细胞转染技术及RNA干扰技术,分别建立和筛选出F10基因稳定过表达及沉默的绒癌细胞系JAR。将30只4~5周龄SPF级裸鼠随机均分为JAR F10过表达组、JAR F10沉默组、JAR未处理组(n=10),分别于颈背部皮下接种5×107个F10基因过表达的JAR细胞、F10基因沉默的JAR细胞及未处理的JAR细胞。接种后每3~4d称重并观察,记录肿瘤发生时间,观察皮下肿瘤的生长情况。绘制在体肿瘤生长曲线,计算各组裸鼠的成瘤率。细胞接种5周后处死小鼠,取瘤组织称重,比较各组的瘤体重量。结果 3组的成瘤率均为100%(10/10)。JAR F10过表达组、JAR F10沉默组、JAR未处理组的成瘤时间分别为4.4±1.1、4.4±1.1、4.6±1.3d,组间比较差异无统计学意义(F=0.097,P=0.908)。与JAR未处理组相比,JAR F10过表达组肿瘤的在体生长速度明显增快,JAR F10沉默组则明显减慢,差异均有统计学意义(P〈0.05)。JAR F10过表达组、JAR F10沉默组、JAR未处理组肿瘤重量分别为607.49±216.19、270.73±81.53、423.87±74.75mg,组间比较差异有统计学意义(F=14.462,P=0.000)。结论 F10基因与绒癌细胞系JAR的增殖调节有关,可增强JAR细胞系在裸鼠体内的致瘤性。Objective To explore the role of the novel hydatidiform mole-related gene F10 in the tumorigenicity of choriocarcinoma cell line JAR in nude mice. Methods By cell transfection and RNA interference, stable F10 gene overexpression and F10 gene-silenced JAR choriocarcinoma cell line were engendered. Thirty SPF nude mice(aged 4-5 weeks) were randomly assigned into 3 groups(10 each): F10 overexpression group(inoculated with F10 gene-overexpressed JAR cells), control group(inoculated with non-treated JAR cells) and F10 gene-silenced group(inoculated with F10 gene-silenced JAR cells). JAR cell suspension(0.2ml with 5×107 cells in each mouse) were injected into the subcutaneous tissue of the back of mice neck. After injection, the conditions of the mice were obser ved, and they were weighed once ever y 3-4 days. The tumor formation time was recorded, the tumor grow th cur ve was plotted, and tumor formation rate was calculated. The mice were sacrificed, and the subcutaneous tumor mass was weighed 5 weeks after the JAR cell injection. Results The rate of tumor formation was 100%(10/10). There was no significant difference in tumor formation time(F=0.097, P=0.908) among the groups of F10 overexpression(4.4±1.1d), F10 silenced(4.4±1.1d), and control(4.6±1.3d). The grow th rate of subcutaneous tumor in F10 overexpression group was significantly higher compared with groups of control and F10 silenced(P〈0.05), and significant difference in tumor growth rate was found between control group and F10 silenced-expression group(P〈0.05). The weight of tumor tissue was significantly different(F=14.462, P=0.000) among the groups of F10 overexpression(607.49±216.19mg), F10 silenced(270.73±81.53mg) and control(423.87±74.75mg). Conclusion F10 gene is involved in the proliferation of JAR choriocarcinoma cell line, and it might enhance the tumorigenicity of JAR cell line in nude mice.
