摘  要:目的:研究黑色素瘤组织中核糖核酸酶抑制因子(RI)的表达量及过表达RI对B16-F10细胞体内外生长的调节作用。方法:取黑色素瘤组织和肿瘤旁正常组织,检测RI的表达量;培养B16-F10细胞,过表达RI后测定细胞的增殖、侵袭行为以及相关基因的表达量;建立黑色素瘤移植瘤动物模型,称量移植瘤的重量。结果:(1)临床组织标本相关结果:不同TNM分期黑色素瘤组织中RI的表达量均低于肿瘤旁正常组织且TNM分期越高、RI的表达量越低;(2)细胞实验相关结果:过表达RI组细胞的OD值显著低于阴性对照组,发生侵袭的细胞数目少于阴性对照组,PKB、GSK-3β、ILK、PI3K、AKT的mRNA含量显著低于阴性对照组;(3)动物实验相关结果:过表达RI组小鼠的黑色素瘤移植瘤重量低于阴性对照组。结论:黑色素瘤组织中RI的表达量显著减少,增加RI的表达能够抑制黑色素瘤细胞的体内外生长。Objective: To investigate the expression of ribonuclease inhibitor (RI) in melanoma tissue and the regulating effects of the over-expressed RI on B16-F10 cell growth in vivo and in vitro. Methods: Melanoma tissue and adjacent normal tissue were collected to detect RI expression levels~ B16-F10 cells were cultured and after overexpression of RI, cell prolifera- tion and invasion as well as related gene expression levels were detected~ animal models with melanoma xenografts were built and weight of xenografts was weighed. Results: (1) Clinical tissue sample-related results: RI expression levels in melanoma tissue with different TNM stages were lower than those in adjacent normal tissue, and the higher the TNM stage, the lower the RI expression levels were; (2) cell experiment-related results: OD value of overexpressed RI group was significantly lower than that of negative control group, the number of invasive cells was less than that of negative control group, and mRNA con- tents of PKB, GSK-313, ILK, PI3K and AKT were significantly lower than those of negative control group;(3) Animal experi ment-related results : weight of melanoma xenografts of overexpression RI group was lower than that of negative control group. Conclusion: RI expression significantly decreases in melanoma tissue, and increasing RI expression can inhibit melanoma cell growth in vivo and in vitro.
