摘 要:目的探讨甲状腺乳头状癌(Papillary Thyroid Carcinomas,PTC)STAT3及其磷酸化水平与三叶因子3(Trefoil Factor 3,TFF3)表达及相关性。方法采用免疫组织化学检测31例组织芯片中PTC及癌旁组织STAT3、p STAT3和TFF3阳性率与平均光密度(AOD)值;Western Blot检测6例PTC和癌旁手术标本中相应蛋白的含量,并进行相关分析。结果 (1)免疫组化:TFF3蛋白在PTC中均高表达(P〈0.001),TFF3阳性率与AOD值在直径〉2 cm组、淋巴结转移组和临床Ⅲ-Ⅳ病例明显高于直径≤2 cm组、无淋巴结转移组和Ⅰ-Ⅱ期者(χ^2=12.120,P=0.001,χ^2=7.517,P=0.043,χ^2=10.490,P=0.003;AOD值:t=2.810,P=0.0064,t=2.234,P=0.039,t=2.926,P=0.0058);PTC中STAT3阳性率和AOD值与临床资料无关。p STAT3为胞核阳性,阳性率和AOD值在PTC组织低于癌旁组织,直径〉2 cm组、淋巴结转移组和临床Ⅲ-Ⅳ病例明显低于直径≤2 cm组、无淋巴结转移组和Ⅰ-Ⅱ期者(χ-2=4.963,P=0.046,χ-2=4.882,P=0.033;AOD值:t=2.195,P=0.041,t=2.790,P=0.0068,t=2.018,P=0.042);TFF3与STAT3呈正相关,与p STAT3呈负相关(P〈0.01)。(2)Western Blot:TFF3和STAT3蛋白在PTC的表达高于癌旁组织(t=2.841,P=0.008,t=2.925,P=0.007),但p STAT3却明显低于癌旁组织(t=2.713,P=0.009)。结论甲状腺乳头状癌TFF3和STAT3高表达、p STAT3低表达对PTC不良预后的预测有指导意义。Objective To investigate the expression and correlation of STAT3, p STAT3 with TFF3 in the papillary thyroid carcinomas(PTC). Methods Immunohistochemistry was used to investigate the positive expression and the average optical density(AOD) value of STAT3, p STAT3 and TFF3 among 31 cases PTC and para-carcinoma tissue chips. Western blot was used to detect the protein content among 6 cases PTC and para-carcinoma specimens. Results(1) Immunohistochemistry: TFF3 was highly expressed in PTC. It's positive rate and AOD value were higher in diameter 2cm, lymph node metastasis group and clinical Ⅲ-Ⅳstage cases than those of diameter ≤2cm group, without lymph node metastasis group and Ⅰ-Ⅱstage(χ^2=12.120, P=0.001,χ^2=7.517, P=0.043, χ^2=10.490, P=0.003; AOD value: t=2.810, P=0.0064, t=2.234, P=0.039, t=2.926, P=0.0058). The positive rate and AOD value of STAT3 had nothing to do with clinical data. p STAT3 mainly expressed in nucleus, whose positive rate and AOD value were lower in para-carcinoma. The positive rates of diameter 2cm group, lymph node metastasis group and clinical Ⅲ-Ⅳstage cases were lower than those of diameter ≤2cm group, without lymph node metastasis group andⅠ-Ⅱstage cases(χ^2=4.963,P=0.046, χ^2=4.882, P=0.033; AOD value: t=2.195, P=0.041, t=2.790, P=0.007, t=2.018, P=0.042). A positive relationship between TFF3 with STAT3, and an inverse relationship between TFF3 with p STAT3 were observed(P0.01).(2) Western blot: The content of TFF3 and STAT3 protein in PTC were higher than para-carcinoma tissue(t=2.841, P=0.008, t=2.925, P=0.007), but p STAT3 was lower than para-carcinoma(t=2.713, P=0.009). Conclusion There may be a guiding significance for poor prognosis prediction of PTC when TFF3 and STAT3 high expressed with low expression of p STAT3 in PTC tissue.
