摘　　要：目的探讨RNAi介导的基因治疗联合天然药物半边旗提取物5F(Ps L 5F)对肿瘤细胞增殖和凋亡的影响。方法采用腺病毒介导Survivin RNAi联合PsL 5F作用乳腺癌细胞MCF-7,免疫印迹法检验Survivin被沉默的程度,通过MTT检测细胞的增殖情况,流式细胞术检测细胞凋亡情况。结果免疫印迹实验发现Survivin蛋白的表达明显被抑制;MTT实验表明联合作用对细胞增殖的抑制作用从单独应用的50.00%(P<0.001),提高到86.12%(P<0.001);流式细胞术也发现联合应用能更大程度地促进细胞的凋亡。结论 Survivin RNAi和PsL 5F联合作用,可显著提高对MCF-7细胞增殖的抑制作用和对细胞凋亡的促进作用,这为肿瘤的综合治疗提供了理论基础和实验室依据。Objective To study the effect of gene therapy,mediated by RNAi combined with PsL 5F on the proliferation and apoptosis of tumor cells. Methods After treated with Survivin RNAi combined with PsL 5F for 48 hours,Survivin was knockdowned by Western blot. The cell proliferation of MCF-7 cells were assayed by MTT,and the flow cytometry was used to detct the cell apoptosis. Results After 48 hours combined treatment,Survivin was significantly knockdowned,and the inhibitory rate on the proliferation of MCF-7 cells improved from about 50 percent（ P〈0. 001） to 86. 12 percent（ P〈0. 001）. And also the combined treatment could strongly promote cells apoptosis. Conclusion Survivin RNAi joint Ps L 5F could enhance the inhibition of MCF-7 cell proliferation and promote cell apoptosis,and it could provide laboratory evidence for reducing dosage and side effects of cancer chemotherapy drugs.
