摘  要:利用正交设计法优选石榴皮中总多酚的提取工艺。以没食子酸为对照,福林试剂为显色剂,用紫外一可见分光光度法在760nm处测定总多酚的含量。采用L9(3^4)正交试验设计,以石榴皮中总多酚含量为考察指标,考察乙醇浓度、提取温度、料液比、提取时间4个因素对提取的影响。结果表明:最佳提取工艺为乙醇浓度为40%,提取温度55℃,料液比1:30,提取时间为30min。该工艺经试验重复性好,操作简单,为石榴皮中总多酚的开发提供了参考依据。The extraction technology of total polyphenols in punica gratanum L. was optimized. The ultraviolet spectrophotometry method was used to measure the total polyphenols content in ethanol extract of Punica gratanum L. with different polar solvent extraction, with Folin's reagent as color reagent and the gallic acid as standard substance and the maximum absorption wave-length of 760 nm. By L9(3^4) orthogonal experimental design, with Punica gratanum L. , as index of total polyphenols, ethanol concentration, extraction temperature, extraction time and ethanol dosage on the extraction were investigated. Results showed that the optimum extraction process was 40% ethanol, 55℃, 30 times the amount of reflux extraction and 30 rain. The experimental reproducibility of the process showed the further development of process a better reference value.
