摘 要:目的:观察穴位埋线对肥胖大鼠SOCS-3基因表达的影响,阐明瘦素抵抗可能发生的分子机制。方法:将普通饲料喂养大鼠作为正常组,挑选经高脂饲料喂养12周体质量超过正常大鼠20%者,随机分模型组和埋线组。埋线治疗后,使用ELISA和RT-PCR等方法观察穴位埋线对各组肥胖大鼠下丘脑胰岛素、瘦素含量、瘦素受体基因(OB-Rbm RNA)表达,观察下丘脑细胞信号转导抑制因子-3(SOCS-3)基因表达;观察外周血清SOCS-3的含量表达。结果:埋线后大鼠血清瘦素和胰岛素含量较模型组肥胖大鼠明显降低(P<0.05),而下丘脑瘦素和胰岛素含量和脑、血瘦素和胰岛素之比较模型组肥胖大鼠均明显升高(P<0.05);埋线后大鼠下丘脑OB-Rbm RNA表达水平与模型组肥胖大鼠比较显著增加(P<0.05),而SOCS-3m RNA表达水平及血清SOCS-3含量明显减少(P<0.05)。结论:穴位埋线改善胰岛素、瘦素血脑转运屏障及胰岛素、瘦素受体后信号传导障碍,进而纠正胰岛素抵抗和瘦素抵抗,达到减肥的目的。Objectives: To observe the effects of SOCS- 3 gene expression on obese rats with catgut implantation at acupoints and clarify the mechanism of leptin resistance( LR) which may be the molecular mechanism. Methods: The author categorized rats fed by standard diet as the normal group and divided experimental obese rats( who were fed by high fat diet for more than 12 weeks and whose weights exceeded 20% of those fed by standard diet) randomly into the model group and catgut implanting group. The author observed the effects of catgut implantation at acupoints on obese rats' hypothalamic insulin( INS),LP content,LP receptor gene( OB- Rbm RNA) expression,hypothalamic suppressor of cytokine signaling( SOCS- 3) expression and content of peripheral serum( SOCS- 3) by means of ELISA and RT- PCR. Results: After the treatment of catgut implantation at acupoints,the contents of serum leptin and of the rats in catgut implanting group decreased significantly compared with those in model group( P 〈0. 05) while the contents of hypothalamus LP and and the ratio increased significantly compared with those in the model group( P 〈0. 05). The rats' hypothalamus OB- Rbm RNA expression level in the catgut implanting group increased significantly compared with those in the model group rats( P 〈0. 05) while the SOCS- 3m RNA expression level and SOCS- 3 content in serum decreased significantly( P 〈0. 05). Conclusions: The study shows that the weight loss mechanism of catgut implantation at acupoints can improve,LP blood brain transport barrier and,LP receptor signal transduction disorder and correct resistance( IR) and LR and help to achieve the purpose of weight loss.
