益气养阴方联合化疗抑制Lewis肺癌移植小鼠肿瘤生长的疗效与机制

摘　　要：目的:观察益气养阴方联合化疗对Lewis肺癌荷瘤小鼠的抑瘤作用,并从CD+4CD+25Treg细胞及Foxp3m RNA表达、IL-2等变化探讨其抗肿瘤机制;方法:近交系雄性C57BL/6纯系小鼠,64只,随机分为对照组、联合组、益气养阴组、化疗组4组,对照组只行造模观察,联合组以化疗联合益气养阴方治疗,益气养阴组只行中药治疗,化疗组只行化疗,观察各组肿瘤体积及抑瘤率,并以ELISA法测定各组血浆IL-2、IL-4、IL-10和IFN-γ浓度,以流式细胞术检测脾脏CD+4CD+25Treg细胞数量,以RT-PCR法测定各组小鼠脾脏Foxp3 m RNA表达;结果:联合组肿瘤体积缩小最明显,抑瘤率最高,DDP组、肺积方组体积缩小、抑瘤率相当;在给药第10、21天,DDP组、肺积方组、联合组血浆IL-2、INF-γ水平均较对照组有差异(P<0.05),其中DDP组较对照组降低(P<0.05),而联合组较对照组升高(P<0.05),DDP组较益气养阴组、联合组均降低明显(P<0.05),联合组较益气养阴组升高(P<0.05);在给药第10天,DDP组、肺积方组、联合组血浆IL-4、IL-10水平均较对照组有差异(P<0.05),其中联合组降低较DDP组、益气养阴组降低有差异(P<0.05),益气养阴组较DDP组降低有统计学差异(P<0.05);第21天DDP组、肺积方组、联合组血浆IL-4、IL-10水平均较对照组降低,降低有差异(P<0.05),但益气养阴组与DDP组比较无差异(P>0.05);在脾脏CD+4CD+25调节性T细胞比例比较中,在第10天,联合组CD+4CD+25调节性T细胞比例较对照组、DDP组、益气养阴组低下(P<0.05),而DDP组与益气养阴组CD+4CD+25调节性T细胞比例无统计学差异(P>0.05),在第21天,联合组CD+4CD+25调节性T细胞比例仍较DDP组、对照组、益气养阴组降低(P<0.05),益气养阴组较DDP组降低(P<0.05);益气养阴组、联合组组内比较,CD+4CD+25调节性T细胞比例无统计学差异(P>0.05),DDP组组内比较,CD+4CD+25调节性T细胞比例第21天较第10天升高(P<0.05);小鼠脾脏中Foxp3 m RNA表达情况:在第Objective： To observe the antitumor effects of Yiqi Yangyin prescription accompanied by chemotherapy and partly demonstrate the mechanism by the varies in CD＋4CD＋25Tregs,mR NA expression of transcription factor Foxp3 and IL- 2,IL- 4,IL- 10 and INF- γ. Methods： Sixty- four C57 BL /6 male mice were randomly divided into four groups： DDP group,comprehensive group,control group,Yiqi Yangyin prescription group. The mice in DDP group were only treated by DDP and those in Yiqi Yangyin prescription group treated only by Yiqi Yangyin prescription and those in control group were only observed and those in comprehensive group were treated by Yiqi Yangyin prescription and DDP. We observed the volume of the tumor and tumor inhibition rate in each group and at the same time contrasted varies of IL- 2,IL- 4,IL- 10 and IFN- γ measured by ELISA and CD＋4CD＋25Treg cell ratio was measured by flow cytometry and mR NA expression of transcription factor Foxp3 measured by reverse transcription- polymerase chain reaction in each group. Result： The volume reduction of the tumor and the tumor inhibitory rate in comprehensive group were the most highest among groups（ P 〈0. 05）. The volume reduction of the tumor and the tumor inhibitory rate were not different between DDP group and Yiqi Yangyin prescription group. The levels of interleukin（ IL）- 2,IL- 4,IL- 10 and interferon（ INF）- γ in plasma of DDP group,comprehensive group and Yiqi Yangyin prescription group were different from those of control group（ P 〈0. 05） and the levels of IL- 2 and IFN- γ in DDP group were lower than that in other groups（ P 〈0. 05） and the levels of IL- 2 and IFN- γ in comprehensive group were higher than those in Yiqi Yangyin prescription group（ P 〈0. 05）. The levels of IL- 4 and IL- 10 in DDP group,Yiqi Yangyin prescription group and comprehensive group were different from those of control group（ P 〈0. 05） after medicine intervention for 10 days.
