同种螺旋神经节和蜗神经抗原免疫致豚鼠听神经病样病变的抗原成分分析

摘  要:用同种螺旋神经节和蜗神经抗原免疫19只豚鼠,依据听觉电生理测试和DP OAE试验结果,判断听神经病豚鼠模型。采用豚鼠血清和Western blotting试验方法,比对分析模型豚鼠与非模型豚鼠针对组织抗原中不同蛋白条带抗体反应情况,探寻可能的致病抗原成分。结果表明:6只豚鼠出现听神经病样表现,Western blotting结果均出现针对相对分子质量3.1×10^4蛋白条带阳性反应。内耳病理观察亦显示其主要免疫炎性损伤部位位于螺旋神经节。实验提示相对分子质量3.1×10^4蛋白成分可能是导致自身免疫性听神经病的主要内耳组织抗原成分。19 guinea pigs were immunized with allogenic spiral ganglion and cochlear nerve antigens, and auditory neuropathy (AN) animal models were evaluated based on the parameters of auditory electrophysiological and distortion product otoacoustic emission (DP OAE ) tests. Western blotting test was performed with animal serum, and the different antibodies against corresponding ingredient of spiral ganglion and cochlear nerve antigens were compared between animal models and controls. The pathogenic composition of antigens could be found. Results showed that AN disorder occurred in 6 guinea pigs, and all of them appeared the positive immune response against relative molecular mass 3. l x 104 protein band appeared in the Western blotting test. The histopathologieal observation of inner ear also showed that the main immune inflammatory injury was located in spiral ganglion, and that relative molecular mass 3.1 × 104 protein was main composition of antigens in the inner ear tissue associated with autoimmune AN. The result would provide the possible assessment for preparing AN animal model and clinical diagnosing autoimmune AN.
