摘 要:目的利用基于L5178Y细胞的涵盖多个遗传学终点的组合试验对土荆芥挥发油的遗传毒性进行评价,为其安全性评价提供理论依据。方法土荆芥挥发油浓度设置为体积分数(v/v)0.001%、0.002%、0.004%和0.008%,对L5178Y细胞进行染毒处理,1%二甲基亚砜(DMSO)为溶剂对照,分别采用小鼠淋巴瘤胸腺嘧啶激酶基因突变试验(MLA)检测TK基因位点突变频率,微核试验(MN)评价细胞染色体损伤,彗星试验检测DNA损伤。结果随土荆芥挥发油剂量的增加,各剂量组L5178Y细胞相对存活率(RS%)下降;MLA试验中TK基因突变频率(TMF)随土荆芥挥发油浓度增加则呈上升趋势(F=2596.8,P=0.00)。TK基因突变频率在土荆芥挥发油各浓度间以及溶剂对照组间的多重比较差异均有统计学意义(P<0.05);微核试验中,各剂量组微核率高于阴性对照组(P<0.05);彗星试验中,除了0.001%组之外,各试验组的彗星尾DNA含量(%)、尾长度和尾矩均高于阴性对照组(P<0.05)。结论土荆芥挥发油可诱导L5178Y细胞基因突变、染色体断裂和DNA损伤,有一定的致突变性。Objective Genotoxicity of volatile oil from Chenopodium ambrosioides L. was evaluated using multi-endpoint genotoxic assay based on Mouse Lymphoma cells L5178 Y to provide theoretical information for its safety evaluation. Methods L5178 Y cells were exposed to volatile oil of C. ambrosioides at volume fraction concentrations of 0. 001%,0. 002%,0. 004% and 0. 008%. 1% dimethyl sulfoxide( DMSO) was used as solvent control. Mouse lymphoma thymidine kinase gene mutation assay( MLA) was used to detect TK gene mutation frequency,micronucleus test( MN) was used to evaluate chromosome cleavage and comet assay to detect DNA damage. Results MLA showed that relative survival( RS%) of L5178 Y cells decreased while TK gene mutation frequency( TMF) increased with the concentration of volatile oil rising compared with the solvent control( F = 2596. 8,P = 0. 00) MN test showed that micronucleus frequency of each treated group was higher than that of the solvent control( P〈0. 05); comet assay results showed that except the 0. 001% treated group,comet Tail DNA%,Tail Length and Tail Moment of the other treated group were higher than the those of solvent control( P〈0. 05).Conclusion C. ambrosioides volatile oil can induce gene mutations,chromosomal breakage and DNA damage in L5178 Y cells,which indicate that the oil may have certain mutagenic effect.
