摘  要:目的通过检测瘤组织BCL-2和BAX蛋白的变化,初步探讨卡铂缓释微球在大鼠皮下移植瘤模型中诱导肿瘤细胞凋亡的作用机制。方法将90只成年SD大鼠建立皮下移植瘤模型并将其随机分为空白对照组、生理盐水组和卡铂缓释微球组,每组各30只。按照给药时间分为0、3、7、14、21天5个观察时间点,在每一时间点取三组大鼠瘤组织进行TUNEL染色检测细胞凋亡以及免疫组织化学染色检测BCL-2和BAX蛋白的含量。结果开始给药时三组大鼠肿瘤细胞凋亡指数比较无显著差异(P>0.05);给药后3、7、14、21天,与空白对照组及生理盐水组相比,卡铂缓释微球组大鼠肿瘤细凋亡指数明显升高(P<0.05)。开始给药时三组大鼠瘤组织BCL-2和BAX蛋白阳性率比较无显著差异(P>0.05);给药后3、7、14、21天,与空白对照组及生理盐水组相比,卡铂缓释微球组大鼠BCL-2蛋白阳性率明显下降,BAX蛋白阳性率明显升高(P<0.05)。结论卡铂缓释微球靶向给药可以增加肿瘤组织内BAX蛋白的表达,减少BCL-2蛋白的表达,诱导肿瘤细胞凋亡。Objective To investigate mechanism of action of slow-released carboplatin microspheres on inducing tumor cells apoptosis in rats copied with subcutaneous transplantation tumor model by detecting the expression of protein BCL-2 and BAX. Method 90 SD rats transplanted with tumor subcutaneously were randomly divided into blank control group, normal saline group and slow-released carboplatin microspheres group, 30 rats in each group. In accordance with the treatment time, there were 5 observation time, including day 0, 3, 7, 12 and 21. Tumor tissue were obtained and stained with TUNEL for detecting the apoptosis at the point of 5 observation time. The level of BCL-2 and BAX proteins were detected through immunohistochemistry staining. Result At day 0, the apoptosis index in each group had no significant difference(P〈0.05). At day 3, 7, 14 and 21, compared with blank control group and normal saline group, apoptosis index of slow-released carboplatin microspheres group increased obviously(P〈0.05). At day 0, the level of BCL-2 and BAX proteins in each group had no significant difference(P〉0.05). At day 3, 7, 14 and 21, compared with blank control group and normal saline group, the level of BCL-2 protein in slow-released carboplatin microspheres group decreased obviously, but the level of BAX protein increased markedly(P〈0.05). Conclusion Slowreleased carboplatin microspheres can induce the apoptosis of tumor cells by increasing the expression of BAX protein and suppressing the expression of BCL-2 protein.
