宫粉羊蹄甲花粉离体萌发培养基的筛选

摘　　要：【目的】筛选最适宫粉羊蹄甲花粉离体萌发的培养基及培养条件,为其花粉活力检测、品种繁育及苗木生产提供参考依据。【方法】宫粉羊蹄甲花粉粒采用液态培养法,设置培养基组分、p H和温度等8个影响因子的梯度值进行逐步迭代优化,以缺素培养为对照(CK),探讨其花粉萌发效果。【结果】培养基各组分对宫粉羊蹄甲花粉离体萌发的影响效果迥异,其中H3BO3对花粉萌发起决定性作用,花粉萌发对蔗糖较敏感。宫粉羊蹄甲花粉在添加8.0%蔗糖、10.0%聚乙二醇-6000(PEG-6000)、0.0020%H3BO3、0.02%Ca Cl2·2H2O、0.02%Mg SO4·7H2O及0.010%KNO3的液体培养基中,于p H 6.8、恒温29℃条件下培养约20 min可观察到最佳萌发效果,萌发率均值达91.230%。【结论】宫粉羊蹄甲花粉在液体培养基各组分及培养条件最佳组合中离体培养才能提高萌发率,花粉管才能正常生长。[Objective ]The optimal medium and culture condition for in vitro pollen germination of Bauhinia variegate L. were screened, in order to provide reference basis for pollen viability determination, varieties breeding and nursery stock production. [Method]The pollen grains were cultivated with liquid medium. And 8 factors affecting pollen germina- tion were selected, including medium composition, pH and temperature etc. And the gradient values of 8 factors were set by iterative optimization method. Then the in vitro pollen germination effect was compared with germination effect of pollen in medium lacking nutritional elements（as control）. [Result]The results showed that there were large differences between effects of medium composition on in vitro pollen germination of B. variegate L.. Most of all, HaBO3 played the decisive role in pollen germination, and the germinating pollen was more sensitive to sucrose. When B. variegate L. in vitro pollen were cultured in the liquid medium supplemented with 8.0% sucrose, 10.0% PEG-6000 （polyethylene glycol-6000）, 0.0020% H3B03, 0.02% CaC12.2H20, 0.02% MgSO4-7H2O and 0.010% KNO3 0.02% at 29℃ and pH 6.8 for 20 min, the optimal germination effect was obtained, the average germination rate reached up to 91.230%. [Conclusion ]The optimal medium and culture condition for in vitro pollen germination of B. variegate L. are helpful to improve pollen germination rate and promote normal growth of pollen tube.
