摘  要:目的观察牙本质基质蛋白1(DMP1)和DMP1环化重组酶(DMP1-Cre)表达的时空特点。方法免疫组织化学染色观察胚胎晚期(E18.5)、1、3周C57BL/6J小鼠DMP1的表达,β-半乳糖苷酶(β—gal)染色观察1、3周DMP1-Cre,ROSA26(ROSA26为Cre报告基因)小鼠中DMP1-Cre表达的时间和空间特点。结果DMP1在小鼠正常发育中,早期无明显表达,在随后的生长发育中,DMP1集中表达于皮质骨中的骨细胞,阳性细胞计数:骨细胞(35±3)个、成骨细胞(10±1)个、软骨细胞(5±1)个,与其他细胞中表达比较差异有统计学意义(P〈0.01),在成骨细胞、软骨细胞及生长板中也有少量表达;DMP1-Cre在骨细胞集中表达,阳性细胞计数:骨细胞(18±2)个、成骨细胞(5±1)个、软骨细胞(6±2)个(P〈0.01),且在软骨细胞和成骨细胞中均有少量表达(P〈0.01)。结论DMP1-Cre在骨骼系统中的多种细胞中均有表达,因此在进行相关基因操作时,这一表达特点需要在结果分析中加以考虑。Objective To observe the expression pattern of dentin matrix proteinl ( DMP1 ) as a promoter of Cre/loxP systems spatially and temporally. Methods El8.5, 1,3 weeks old C57BL/6J mice and 4 weeks DMP1 - Cre ; ROSA 26 genotype mice were bred. Immumohistochemical staining and 13 - galactosidase (β- gal) staining were employed to observe the expression pattern of DMP1 and DMP1 - Cre ; ROSA 26 mice. Results DMP1 showed no observable expression in late embryo stage of C57BL/6J mice. However, in early growth stage, DMP1 was mainly expressed in osteocytes of cortical bones (compared with other cells, immuohistochemical staining positive cells : osteocytes 35 ±3, osteoblasts 10 ±1, chondrocytes 5 ±1, P 〈0. 01 ; n =5/group), and small amounts were found in osteoblasts, chondrocytes, and the growth plate; DMP1 - Cre was found assembled in osteocytes (immuohistochemical staining positive cells : osteocytes 18 ±2, osteoblasts 5 ±1 ,chondrocytes 6 ±2 ; P 〈 0. 01 ), and a few in osteoblasts, chon- drocytes (P 〈 0. 01 ) in DMP1 - Cre ; ROSA 26 mice. Conclusion DMP1 - Cre expressed in many kinds of cells of skeletal system. More attention should be paid when it was used in relative gene manipulations and this effect should be taken into consideration in the analysis to the outcomes.
