摘 要:目的在体外构建经骨保护素(OPG)共价修饰的去细胞猪主动脉瓣,观察复合瓣膜对脐静脉内皮细胞增殖和迁移的影响。方法首先应用聚乙二醇(PEG)对去细胞主动脉瓣进行PEG化,再通过迈克尔加成反应将OPG共价修饰到去细胞瓣上。免疫荧光检测OPG共价修饰去细胞瓣的效果;细胞毒性实验测试复合瓣膜对脐静脉内皮细胞的毒性,观察复合瓣膜对脐静脉内皮细胞增殖和迁移的影响。结果免疫荧光证实OPG成功修饰到去细胞瓣上,复合瓣膜的细胞增殖率在24、48h分别为(102.63±3.08)%和(105.77±4.19)%,且复合瓣膜上内皮细胞数多于其余2组瓣膜,且迁移实验显示复合瓣膜组迁移的细胞数也多于其余2组瓣膜。结论成功制备经OPG修饰的去细胞瓣,且复合瓣膜可促进脐静脉内皮细胞的增殖和迁移。Objective To eslablish PEGylated decellularized valves modified by osteoprotegerin (OPG) in vitro, and explore their influences on human umbilical vein endothelial cells (HUVECs) proliferation and migration. Methods First, 4 - arm - PEG - acrylate was used to PEGylate the decellularized porcine aortic valves. Second, we established PEGylated decellularized valves modified by OPG in vitro via Michael addition reaction. The establishment result of composite valves was observed by immunofluorescence. The toxicity on HUVECs was detected by cytotoxicity test. The influences of composite valves on HUVECs proliferation and migration were analyzed. Results We confirmed that PEGylated decellularized valves were successfully modified by OPG via immunofluorescence. The relative growth rate of composite valves at 24 h, and 48 h was ( 102. 63 ±3.08) % and ( 105.77 ±4. 19) % respectively, and the number of proliferating cells on the composite valves outnumbered that on other valves. Migration test indicated that the migratory cells induced by composite wlves also outnumbered that on other valves. Conclusion We successfully established PEGylated decellularized valves modified by OPG, and they could promote HUVECs proliferation and migration in vitro.
