摘 要:作者运用分子生物学方法,制备了生物素标记人乳头状瘤病毒(HPV)16型DNA探针,用斑点杂交检测宫颈癌组织DNA中HPV 16 DNA相关序列。在28例宫颈癌组织中,有16例呈HPV 16 DNA杂交阳性,阳性率为57%;4例正常宫颈组织呈阴性反应;4例宫颈慢性炎症中有1例呈阳性反应。实验阳性对照HPV 16质粒DNA呈杂交强阳性,以λ-噬菌体DNA及NIH/3T3小鼠成纤维细胞DNA为实验阴性对照,均呈杂交阴性。结果证明,HPV的基因组存在于宫颈癌组织中,提示HPV的感染与宫颈癌的发生有关。本实验采用生物素标记HPV 16 DNA探针,检测特异性强,方法简便,为开展宫颈癌病毒病因的普查及其研究,提供了一个快速敏感的方法。Biotin-labelled human papillomavirus(HPV)_(16) type DNA probe was prepared bythe techniques of molecular biology. Anddot hybridization technique was used todetect the HPV 16 homologous sequencesin the tissues DNA of human cervical car-cinoma. The results indicated that 16 casesout of 28 of the human cervical carcinomatissues were positive. The positive ratewas 57%.The other 4 cases of normal uter-ine cervix tissues were negative. Only 1in 4 chronic cervicitis tissues showed posi-tive. The HPV 16 plasmid DNA, as thepositive control group, showed strong posi-tive, while λ-phage DNA was negative. Therusults have shown that the genome of theHPV actually exists in the tissue of thecervical carcinoma and that there is a closerelationship between the cervical carcinomaand HPV infection. This experiment adopt-ed the Biotin-labelled HPV 16 DNA probe.And it may provide us with a quick andsensitive method for investigation of theinfection of HPV and its role in the carci-nogenesis of cervical carcinoma.
