摘  要:间充质干细胞具有自我更新和多向分化的能力,能在体外诱导成脂细胞、软骨细胞、骨细胞和肌细胞等.脂肪间充质干细胞(adipose mesenchymal stem cells,AMSCs)取材方便,来源较为丰富.本试验利用hedgehog信号通路的特异性抑制剂cyclopamine(CPA)对牛AMSCs的细胞分裂周期机制做了初步探讨.先用血清饥饿法使细胞处于G0/G1期,再经CPA处理,检测不同处理时间的细胞周期蛋白cyclinA、cyclinD和cyclinE的表达情况.经CPA处理的细胞,其周期蛋白cyclin A、cyclin D和cyclin E的表达水平显著低于对照组.表明,CPA通过抑制细胞周期蛋白基因的表达从而抑制了细胞的增殖.Mesenchymal stem cells (MSC) have the capability of self-renewal and multilineage differentiation,which could differentiate into many kinds of cells such as adipocytes, chondrocytes, osteoblasts and muscle cells. Adipose mesenehymal stem cells (AMSCs) could easily be obtained and are widely used in cell differentiation experiments. In present study, bovine AMSCs were treated by cyclopamine (CPA),a specific inhibitor of hedgehog signaling pathway,to evaluate its effect on the cell cycle of AMSCs. The cells were firstly synchronized to G0/G1 stage by serum starvation method, the treated by CPA for different time periods. After that,the expression levels of cell cycle related genes, including cyclin A, cyclin D and cyclin E, were examined and compared. The results showed that the expression levels of cyclin A, cyclin D and cyclin E in CPA-treated cells were signif- icantly lower than controls,which indicated that CPA treatment could inhibit the cell proliferation in bovine AMSCs through the suppression of cell cycle raltecl genes expression.
