摘 要:目的总结国内外对Musashi homolog 1(简称Msi1)与各种恶性肿瘤发生发展的关系及作用机制的研究现状。方法应用PubMed及CNKI期刊全文数据库检索系统,检索2010-06-2015-6文献,以"Msi1、MSI-1、Musashi-1和肿瘤"等为关键词,共检索到英文文献211条,中文文献180条。纳入标准:1)Msi1的研究进程;2)Msi1与肿瘤之间的关系;3)发表年限较近的文献。剔除标准:1)与肿瘤不相关;2)年代久远;3)重复性。根据上述标准纳入分析文献33篇。结果 Msi1是通过调节转录后的翻译过程来保持干细胞处于未分化的状态,通过调节Notch,Wnt/β-catenin信号通路直接或间接地影响干细胞细胞增殖和细胞命运的决定以及肿瘤形成的发展。Msi1基因在各种恶性肿瘤中高表达,其作用是参与影响肿瘤有关信号通路、细胞增殖及其凋亡等;降低Msi1基因的表达水平可有效诱导癌症细胞凋亡,阻止有丝分裂和细胞周期进程,抑制肿瘤细胞增殖并导致肿瘤消退。结论 Msi1通过对多种基因的转录后调控参与了肿瘤的发生发展和转移等过程,且与肿瘤的预后及治疗有关联。Msi1的研究将对临床肿瘤疾病基因层面的深入研究和诊断治疗提供新途径,有望成为肿瘤诊疗及靶向药物研究的新靶点,但仍需要进一步的研究证实。OBJECTIVE To summarize the research status of the relationship and the mechanism of Musashi-1 and the development of a variety of malignancies. METHODS Application of PubMed and CNKI database from retrieval system,retrieving the all English literature with "MSI-1, Msil, Musashi-1 and tumor" as the keywords from June 2010 to June 2015,In total 211 English literature and 180 Chinese literatures were retrieved. Inclusion criteria: 1) Musashi-1 re search process; 2)the relationship between Musashi 1 and tumor; and 3) publication year closer in. Exclusion criteria were: 1) not associated with tumor; 2) old years ; 3) repeatability. Finally 33 documents were inclueded in the analysis based on the above criteria. RESULTS Musashi-1 is forming stem ceils in an undifferentiated state,by regulating Notch, Wnt/β catenin signaling pathway directly or indirectly to affect the stem cell proliferation, differentiation and tumor formation. Musashi-1 gene is highly expressed in a variety of malignant tumors,its role is involved in influencing many aspects of cancer related signaling pathways, cell proliferation and apoptosis; downregulation of Musashi-1 gene can effectively induce apoptosis in cancer cells, prevent mitosis and cell cycle progression, inhibit of tumor cell proliferation and lead to tumor regression. CONCLUSIONS By post-transcriptional regulation of multiple genes Musashi-1 involved in the process of tumor growth and metastasis as well as the prognosis and treatment of related cancer. Further study of Musashi-1 will be necessary for the clinical diagnosis and treatment of neoplastic disease,It is expected to become a new target for cancer diagnosis and targeted drug research.
