摘  要:目的探索咯利普兰对大鼠脊髓横断损伤后神经元凋亡的影响。方法取成年SD大鼠60只分为假手术组(Sham组)、损伤组(SCI组)及咯利普兰治疗组(R组)各20只。在R组及SCI组建立大鼠脊髓横断损伤模型,Sham组大鼠仅打开椎板;R组术后立刻皮下注射咯利普兰,SCI组及Sham组皮下注射等体积二甲基亚砜。于术后6 h、12 h、24 h、72 h检测各组脊髓半胱氨酸天门冬氨酸蛋白酶3(caspase-3)及B淋巴细胞瘤蛋白2(Bcl-2)的表达水平,并计算各组脊髓的脱氧核苷酸末端转移酶介导的d UTP缺口末端标记(TUNEL)染色阳性细胞数量。结果术后各时间点,R组的Bcl-2表达水平高于SCI组及Sham组(P<0.05),R组的caspase-3表达水平低于SCI组(P<0.05);术后12 h、24 h、72 h,R组TUNEL阳性细胞数量少于SCI组(P<0.05)。结论咯利普兰可明显减少大鼠脊髓横断损伤后神经元的凋亡。Objective To investigate the effect of rolipram on neuronal apoptosis after spinal cord transverse injury in rats. Methods A total of 60 adult SD rats were divided into sham-operation group (Sham group, n = 20), spinal cord injury group( SCI group, n = 20) and rolipram treatment group( Group R,n = 20). The rat model of spinal cord transverse injury was established in SCI group and Group R,and the rats in Sham group were only disposed of opening the lamina. Group R was injected subcutaneously with rolipram immediately after operation, and SCI group and Sham group were injected subcutaneously with the same volume of dimethyl sulfoxide. The expression levels of spinal cord caspase-3 and Bcl-2 were detected in each group on the 6th,12th,24th,and 72nd hour after operation. The amounts of [ terminal deoxynucle- otidyl transferase(TdT)-mediated dUTP nick end labeling,TUNEL] positive cells in spinal cord were counted in each group. Results At each time point after operation, the expression level of Bcl-2 in Group R was significantly higher than that in SCI group or Sham group (P 〈 0.05 ), and the expression level of caspase-3 in Group R was lower than that in SCI group (P 〈 0.05 ). On the 12th ,24th,and 72nd hour after operation,the number of TUNEL positive cells in Group R was significantly lower than that in SCI group (P 〈 0.05 ). Conclusion Rolipram can significantly reduce the neuronal apoptosis after spinal cord transverse injury in rats.
