摘  要:目的研究6个不同城市大气污染颗粒PM_(2.5)对肺癌A549细胞炎症因子IL-1β的影响,探讨PM_(2.5)成分和炎症因子IL-1β之间的关系。方法采集湛江、广州、武汉、上海、北京和成都6个城市PM_(2.5),测定不同城市PM_(2.5)中金属元素及多环芳烃(PAHs)的含量。将不同城市PM_(2.5)以100μg/m L染毒A549细胞24 h,荧光定量PCR检测不同城市PM_(2.5)染毒细胞后细胞内炎症因子IL-1β基因表达水平。通过线性回归和相关性分析研究PM_(2.5)成分和细胞炎症因子之间的关系。结果 6个城市PM_(2.5)中均含有高浓度的PAHs,其中广州、武汉、上海3个城市PM_(2.5)中PAHs浓度高达900 pg/μg,高于其他3个城市。6个城市PM_(2.5)金属成分中,过渡金属元素Fe、Cu、Ni含量较高。6个城市PM_(2.5)均能够引起A549细胞内IL-1β基因表达水平上调,PM_(2.5)中PAHs和Ni元素含量与细胞内IL-1β表达水平呈显著相关。结论不同城市的PM_(2.5)能上调细胞IL-1β的表达,PM_(2.5)上调细胞内IL-1β的作用与PAHs和Ni元素相关。Objectives To investigate the effects of PM_(2.5)on the expression of inflammatory factor IL- 1β in lung cancer cell line( A549 cells) and the associations between PM_(2.5)composition and the expression of IL- 1β.Methods The concentration of metals and PHAs in PM_(2.5) collected from six cities,Zhanjiang,Guangzhou,Wuhan,Shanghai,Beijing and Chengdu were analyzed. A549 cells were exposed to PM_(2.5) at a dose of 100 μg / m L for 24 h;and the expression of IL- 1β in A549 cells was tested by Q-PCR. The association between the composition of PM_(2.5)and the expression of IL- 1β was assessed by using linear regression and correlation analysis. Results The content of PAHs in PM_(2.5) from Guangzhou,Wuhan and Shanghai was about 900 pg / μg,which was higher than that in other three cities. The concentration of Fe,Cu,and Ni was very high in all PM_(2.5) samples,and these transition metals could induce the expression of IL- 1β in A549 cells. High expression of IL- 1β was strongly associated with high concentrations of PAHs and Ni in PM_(2.5). Conclusions PM_(2.5) collected from 6 cities could induce the expression of IL- 1β in A549 cells. The upregulation of IL- 1β in A549 cells induced by PM_(2.5) was associated with the high PAHs and Ni content in PM_(2.5).
