摘  要:为了给烟梗果胶含量的准确测定提供参考依据,对酶解-连续流动法在烟梗果胶含量测定中的适用性进行考察,并采用单因素实验分析法对酶解条件进行优化.结果表明,果胶酶的最优酶解条件为:酶用量15mg、缓冲液体积60mL、缓冲液pH4.0、酶解温度55℃、酶解时间3h;在此条件下,方法的线性相关系数为0.9999,回收率为97.8% ~99.2%,且日内RSD为1.06% ~2.43%,日间RSD为1.87%.该方法检测结果令人满意,能用于烟梗中果胶含量的准确测定.In order to provide a reference for determining pectin in tobacco stem exactly,the applica-bility for the determination of stem pectin content by Hydrolysis - continuous flow method was studied, meanwhile the hydrolysis -continuous flow method was developed,and the enzymatic hydrolysis conditions were optimized for 15 mg of pectinase at 55 ℃ for 3 h in 60mL of sodium citrate /citric acid (pH4.0)buffer solution by single factor experiments during the research.Under this condition,the method has good linearity with a correlation coefficient of 0.999 9,while the recovery of pectin in tobacco stem ranged from 97.8% to 100.3%,and the RSD was 1.06% to 2.43% intraday,1.87% during the day.It showed that the method was satisfactory and could be used for the determination of pectin in tobacco stem simultaneously.
