宫颈癌中异常表达microRNA与HPV-16 E6/E7基因相关性分析

摘　　要：目的探讨宫颈癌组织中差异表达的微小RNA(microRNA,miR)与高危人乳头状瘤病毒(human papillomavirus,HPV)-16感染的关系。方法 20例宫颈癌患者的宫颈癌组织(宫颈癌组)和10例因子宫良性病变切除子宫患者的正常宫颈组织(对照组),采用荧光定量PCR(real-time PCR,RT-PCR)检测2组宫颈组织miR-497、miR-21、miR-9、miR-218、miR-31、miR-195表达情况;采用RT-PCR检测宫颈癌SiHa细胞(HPV-16阳性)和C-33A细胞(HPV-16阴性)中6个miRNAs相对表达量;SiHa细胞分别转染HPV-16E6siRNA和HPV-16E7siRNA,采用RT-PCR检测转染后6个miRNAs相对表达量,并与阴性对照进行比较。结果宫颈癌组miR-218(0.11±0.08)、miR-195(0.20±0.12)和miR-497(0.26±0.10)相对表达量较对照组(均为1)明显下调(P<0.05),miR-21(3.37±0.05)、miR-9(1.97±0.03)、miR-31(7.09±0.11)较对照组(均为1)明显上调(P<0.05);宫颈癌SiHa细胞miR-497(0.70±0.06)、miR-218(0.26±0.03)相对表达量较C-33A细胞(均为1)明显下调(P<0.05),miR-21(1.80±0.03)、miR-9(1.69±0.05)、miR-31(1.85±0.04)相对表达量较C-33A细胞(均为1)明显上调(P<0.05),miR-195相对表达量(0.98±0.03)与C-33A细胞(1)比较差异无统计学意义(P>0.05);转染HPV-16E6siRNA的SiHa细胞中miR-21(0.56±0.20)、miR-9(0.60±0.10)、miR-31(0.59±0.10)相对表达量较阴性对照组(均为1)明显下调(P<0.05),miR-497(1.40±0.12)、miR-218(1.89±0.09)相对表达量较阴性对照组(均为1)明显上调(P<0.05),miR-195相对表达量(0.99±0.02)与阴性对照组(1)比较差异无统计学意义(P>0.05);转染HPV-16 E7siRNA的SiHa细胞中miR-21(0.56±0.10)、miR-9(0.62±0.90)、miR-31(0.65±0.03)相对表达量较阴性对照组(均为1)明显下调(P<0.05),miR-497(1.41±0.15)、miR-218(1.60±0.18)相对表达量较阴性对照组(均为1)明显上调(P<0.05),miR-195相对表达量(0.97±0.04)与阴性对照组(1)比较差异无统计学意义(P>0.05)。结论宫颈癌组织中异常表达的miRNA与HPV-16E6/E7基因有关。Objective To explore the relationship between abnormal expression of microRNA （miR） and human papillomavirus （HPV）-16 E6/E7 genes in patients with cervical cancer. Methods The expressions of miR-497, miR-21, miR-9, miR-218, miR-31 and miR-195 were detected by real time fluorescence quantitative PCR （RT PCR） technique in cervical cancer tissues from 20 patients with cervical cancer （cancer group） and normal cervical tissues from 10 patients undergoing hysterectomy for benign uerine diseases （control group）. The relative expressions of 6 miRNAs were detected in cervical cancer cell line SiHa （HPV-16 positive） and C-33A （HPV-16 negative） by using RT-PCR technique. HPV-16 E6 and HPV-16 E7 siRNA were transferred to SiHa cells. The relative expression of 6 miRNAs was detected by using RT-PCR, and compared with negative controls. Results The relative expressions of miR-218 （0. 11± 0.08）, miR 195 （0.20±0. 12）, miR-497 （0. 26±0. 10） in cancer group were obviously dowmregulated in comparison with those in control group （1, 1, 1） （P±0.05）, and the relative expressions of miR 21 （3.37±0.05）, miR-9 （1. 97±0.03） and miR-31 （7.09±0. 11） in cancer group were obviously up-regulated in comparison with control group （1, 1, 1） （P〈0.05）. The relative expressions of miR-497 （0.70 ± 0.06） and miR-218 （0.26 ± 0.03） were obviously down-regulated （P〈0.05）, and miR-21 （1.80±0.03）, miR-9 （1.69±0.05） and miR-31 （1.85±0.04） were obviously up-regulated in SiHa cells （P〈0.05） in comparison with those in C-33A ceils （1, 1, 1, 1, 1）, and there was no significant difference in the relative expressions of miR-195 between SiHa cells （0. 98 ± 0. 03） and C-33 cells （1） （P〉 0.05）. The relative expressions of miR-21 （0.56±0.20）, miR-9 （0.60±0.10） and miR 31 （0.59±0.10） were significantly down-regulated （P〈0.05） and the relative expressions of miR-497 （1. 40 ± 0. 12） and miR-218 （1. 89 ± 0. 09�
