侧脑室引流术对特定插线深度大脑中动脉阻塞模型大鼠病死率的影响

摘  要:目的探讨降低大脑中动脉阻塞(middle cerebral artery occlusion,MCAO)模型大鼠病死率的有效方法。方法165只大鼠随机分成插线18mm组56只、插线17.3mm组54只和插线17.3mm+引流术组55只。3组均采用改良Longa法制作MCAO模型,分别于≤1d、〉1~3d、〉3~7d时统计大鼠死亡数,比较3组大鼠MCAO模型成功率和各时间点病死率。结果插线18mm组、插线17.3mm组和插线17.3mm+引流术组MCAO模型成功率分别为89.29%、92.59%、90.91%,两两比较差异无统计学意义(P〉0.05);≤1d时,3组大鼠病死率比较差异无统计学意义(P〉0.05);〉1~3d时、〉3~7d时插线18mm组(24.0%、10.0%)和插线17.3mm组(24.0%、6.0%)大鼠病死率高于插线17.3mm+引流术组(10.0%、0)(P〈0.05),插线18mm组与插线17.3mm组比较差异无统计学意义(P〉0.05)。结论侧脑室引流术联合17.3mm插线深度能明显降低MCAO模型大鼠病死率。Objective To explore an effective method to reduce the fatality of middle cerebral artery occlusion (MCAO) in rat models. Methods A total of 165 rats were randomly divided into 18-mm thread group (n=56), 17.3-mm thread group (n= 54) and 17.3-mm thread plus paraventriculostomy group (n = 55). The MCAO models were established by using modified Longa method. The numbers of died rats were calculated respectively at the time points of within 1 day, from 1 to 3 days and from 3 to 7 days, and the success rates and fatalities of MCAO models were compared respectively at each time point among three groups. Results The success rate of MCAO models was 89.29%, 92.59% and 90.91% in 18-mm thread group, 17.3-mm thread group and 17.3-ram thread plus paraventriculostomy group, respectively, showing no significant difference between each two groups (P〉0.05). There were no significant differences in the fatalities among three groups within 1 day (P〉0.05). The fatalities from 1 to 3 days and from 3 to 7 days were significantly higher in 18-mm thread group (24.0%, 10.0%) and 17.3-mm thread group (24.0%, 6.0%) than those in 17.3-mm thread plus paraventriculostomy group (10. 0%, 0) (P〈 0.05), and there were no significant differences between 18-mm and 17.3-mm thread groups (P%〉0.05). Conclusion Paraventriculostomy plus 17. 3-mm thread can obviously reduce the fatality of MCAO rat models.
