摘 要:目的观察间断强刺激、慢性持续性温和刺激对老年大鼠心血管活性物质一氧化氮(NO)、内皮素-1(ET-1)水平的影响。方法 Wistar老年大鼠共24只,随机分为空白对照组、间断强刺激干预组、慢性持续性温和刺激干预组。适应性喂养后,分别对间断强刺激干预组、慢性持续性温和刺激干预组给予间断高强度应激刺激、持续性温和刺激(均包括冰水游泳、热刺激、禁食、禁水、夹尾、昼夜颠倒、高速水平摇晃、频闪灯照射,程度及频次不同)。经观察大鼠体重变化、糖水偏嗜等一般情况;同时检测血管活性物质(NO、ET-1)水平。结果分别给予间断高强度应激刺激、持续性温和刺激56 d后,两种刺激干预组与空白对照组比较,体重上升幅度、糖水偏嗜度均有降低趋势,但差异无统计学意义(P>0.05)。两种刺激干预组与空白对照组比较,慢性持续性温和刺激干预组NO水平明显升高(P<0.05);两种刺激干预组与空白组比较,慢性持续性温和刺激干预组ET-1水平明显升高(P<0.05)。结论不同应激刺激均可导致老年大鼠出现抑郁表现,同时可影响心血管活性物质(NO、ET-1)的表达,可能以此途径影响心血管疾病的发病及预后。Objective To observe the influence of intermittent strong stimulation and chronic persistent mild stimulation on the levels of nitric oxide (NO) and endothelin 1 ( ET-1 ) in senile rats. Methods A total of 24 senile Wistar rats were randomly divided into the control group, continuous strong stimulation intervention group, chronic persistent mild stimula- tion intervention group. After the feeding on adaptability, the continuous strong stimulation intervention group received strong stimulation while the chronic persistent mild stimulation intervention group received the mild stimulation, the stimu- lation in both groups including water swimming, thermal stimulation, fasting, ban water, tail clip, day and night upside down, high-speed horizontal shaking, and flashing light illuminate were performed for 56 days. The change of body weight and preference for sugar were observed ;while the levels of vascular active substances such as NO and ET1 were tested. Results After the stimulation, the weight increase and preference for sugar in both stimulation intervention groups de- creased not obviously as compared with the control group( P 〉 0.05 ). The levels of NO and ET-1 increased obviously in the chronic persistent mild stimulation intervention group when compared with the control group (P 〈 0. 05 ). Conclusion Every kinds of stimulations will cause the depression in the senile rats, and affect the levels of NO and ET- 1 ,which plays an important role in the pathogenesis and prognosis of cardiovascular disease.
