温州地区原发性肝细胞癌TP53基因249密码子突变研究

摘  要:目的检测温州地区原发性肝细胞癌的TP53基因249密码子突变情况,并探讨其在肝细胞癌中所起的作用及其临床意义。方法收集石蜡包埋的原发性肝细胞癌组织120例,应用Sanger测序技术检测突变情况。运用Spearman检验分析肿瘤组织中TP53基因249密码子突变与原发性肝细胞癌患者临床病理特征之间的相关性。结果 TP53基因249密码子在120例肝癌组织中总突变率为3.33%(4/120),进一步分析显示TP53基因249密码子突变与肝细胞癌患者的临床病理因素未发现存在相关性(P>0.05)。结论在温州地区,TP53基因249密码子突变为非高发事件,未发现其与原发性肝细胞癌的进展相关。Objective To investigate the mutation of TP53 gene codon 249 in primary hepatocellular carcinoma (HCC) in Wenzhou area, and explore its role in the development of HCC. Methods A total of 120 paraffinembedded primary HCC tissues were collected, the Sanger sequencing technique were used to detect the mutation. The Spearman test was applied to analysis the correlation of the mutation of TP53 gene codon 249 and the clinicopathological features in HCC patients. Results Mutation of codon 249 was found in 4 cases with a mutation rate of 3.33% (4/120); the research also showed that the mutation of codon 249 had no correlation with the clinicopathological feature in HCC patients (P〉0.05). Conclusion Our data suggests that the incidence of mutation of p53 gene codon 249 in Wenzhou area is low and little contribution to the development of the hepatocelluar carinoma.
