摘 要:目的应用基于核磁共振氢谱(nuclear magnetic resonance ~1H,NMR ~1H)的代谢组学方法研究红景天对慢性冷应激刺激模型大鼠血浆和尿液代谢组的影响。方法雄性Wistar大鼠30只,随机分为3组:正常对照组、模型组和红景天组。模型组和红景天组大鼠每天置于4℃环境中4 h,连续刺激2周,同时红景天组给予红景天水煎液5 g·kg^(-1)·d^(-1)灌胃。正常对照组和模型组给予等体积的纯净水灌胃。采集大鼠血浆和尿液的NMR ~1H谱,运用主成分分析法(principal component analysis,PCA)进行数据分析。结果红景天组与模型组代谢特征明显不同,其代谢特征更接近正常对照组。与模型组比较,红景天组血浆中甘氨酸升高,乳酸、乙酰乙酸、肌酸、肌酐、甘油磷脂胆碱、磷脂胆碱、牛磺酸、氧化三甲胺和甜菜碱降低;尿液中色氨酸增加,α-酮戊二酸、乙酰乙酸、柠檬酸、氧化三甲胺、甜菜碱、牛磺酸和马尿酸减少。结论红景天能够调控机体能量代谢,以及牛磺酸、氨基酸(甘氨酸和色氨酸)和胆碱的代谢,对冷应激状态下代谢紊乱大鼠具有保护作用。Objective With the application of nuclear magnetic resonance ~1H spectroscopy(NMR ~1H),we study the effect of Herba Rhodiolae Sacrae(HRS) decoction on the plasma and urine metabolites of chronic cold stress rats.Methods Thirty male Wistar rats were divided into normal group,model group and HRS group,10 rats in each group. Rats of model group and HRS group were exposed to 4 ℃ for 4 h each day,and the stress continued for two weeks. Simultaneously,HRS group was given intragastric gavage with the decoction of HRS 5 g·kg^(-1)·d^(-1),and normal group and model group were given the equal dose of pure water. Plasma and urine samples were collected when the experiment was finished.~1H NMR spectra of each sample was detected and then was analyzed by principal component analysis(PCA)method. Results Compared with the model group,HRS group had an increase of plasma glycine,and a decrease of plasma lactate,acetoacetate,creatine,creatinine,glycerol phospholipid choline(GPC),phospholipid choline(PC),taurine,trimethylamino oxide(TMAO)and betaine. HRS group also had an increase of urine tryptophan,and a decrease of urine 2-ocoglutarate, acetoacetate, citrate, TMAO, betaine, taurine, and hippuric acid. The changes of plasma and urine metabolites of HRS group were similar to those of the normal group. Conclusion The results of metabonomics study showed that HRS has regulatory effect on energy metabolism,and on the metabolism of taurine,amino acids(glycine and tryptophan) and choline,which elucidates the metabolism regulative mechanism of HRS in protecting rats from chronic cold stress.
