摘 要:目的研究去甲肾上腺素对人咽鳞状细胞癌细胞系FaDu增殖、侵袭及迁移等细胞生物学行为的影响。方法分别采用MTT实验、Transwell细胞侵袭及迁移实验和划痕实验检测不同浓度去甲肾上腺素对人咽鳞状细胞癌细胞系FaDu增殖、侵袭及迁移能力的影响,同时观察普萘洛尔预处理对去甲肾上腺素作用的干预效果。结果 MTT实验、Transwell实验和划痕实验结果表明浓度为10μmo/L的去甲肾上腺素可以显著促进人咽鳞状细胞癌细胞系FaDu的增殖、侵袭及迁移(P<0.001),而浓度为1μmol/L的普萘洛尔预处理可以有效抑制去甲肾上腺素对人咽鳞状细胞癌细胞系FaDu增殖、侵袭及迁移的促进作用(P<0.001)。结论去甲肾上腺素对人咽鳞状细胞癌细胞系FaDu的增殖、侵袭及迁移有促进作用,而非选择性β肾上腺素能受体阻断剂普萘洛尔可以抑制该促进作用。Objective To investigate the effects of norepinephrine( NE) on the biological behavior of human pharyngeal squamous cell carcinoma cell line FaDu. Methods MTT assay was used to evaluate the influence of NE on the proliferation of tumor cells. Transwell invasion assay was used to evaluate the influence of NE on the invasion of tumor cells. The effects of NE on the migration of tumor cells were measured by Transwell migration assay and wound-healing assay. Additionally,the effects of the β-adrenergic antagonist propranolol on the NE-induced proliferation,invasion and migration of tumor cells were measured. Results 10 μmol / L NE promoted the proliferation,invasion and migration of tumor cells significantly( P〈0. 001). However,pretreatment with 1 μmol / L propranolol effectively inhibitedthe NE-induced proliferation,invasion and migration( P〈0. 001). Conclusion NE promotes the proliferation,invasion and migration of human pharyngeal squamous cell carcinoma cells. These effects can be suppressed by propranolol.
