胃癌CyclinD_1及RasP21蛋白的表达与分化程度及预后关系的探讨

摘　　要：目的:应用流式细胞仪检测正常胃组织及胃癌组织中细胞周期素D1及Ras P21蛋白的表达,探讨其与胃癌分化程度及预后的关系。方法:对40例胃癌组织细胞进行细胞周期素D1及Ras P21蛋白含量的定量检测,10例正常胃组织作参照。结果:(1)正常胃组织中细胞周期素D1的无表达,72.5%的胃癌组织细胞周期素D1呈阳性表达,其表达与胃癌恶性程度及预后有明显相关性(P<0.01)。(2)正常胃组织中Ras P21蛋白无表达,62.5%胃癌组织中Ras P21蛋白表达呈阳性;其表达与胃癌恶性程度及预后有明显相关性(P<0.05)。结论:细胞周期素D1及Ras P21蛋白过表达是导致胃癌的发生的重要因素之一,联合检测cyclin D1蛋白及Ras P21蛋白不仅能揭示胃癌的发病机理,同时有利于胃癌病人预后的评估。Objective：To detect the expression of cyclinD1 and RasP21 proteins in normal gastric tissue and gastric cancer,and to study their correlation with differentiation of gastric cancer. Methods：By using Flow cytometry to detect the expression of cyclinD1 and RasP21 proteins in 10 normal gastric tissue and 40 gastric cancer tissue. Results： （ 1 ） The expression of cyclinD1 was negative in normal gastric tissue. 72. 5% gastric cancer tissure were positive,cyclinD1 positive expression rate and content were correlated with differentiation of gastric cancer and rate of 5 year survival. （P〈0.01）. （2）The expression of RasP21 was negative in normal gastric tissue;over-expression was fond in 62 . 5%gastric cancer tissue;RasP21 positive expression rate and content were correlated with differentiation of gastric cancer and rate of 5 year survival. （P〈0. 05）. Conclusion：The overexpression of cyclinD1 and RasP21 proteins are lead to gastric cancer＇s important reason. To detect cyclinD1 and RasP 21 of proteins may find gastric cancer＇s mechanism,and evaluate prognosis of gastric cancer.
