摘  要:目的观察TGF-β1对人绒毛膜癌细胞肿瘤干性标记物CD133表达的促进作用,探讨其对上皮间质转化(EMT)相关标志物基因表达及迁移能力的影响。方法体外培养人绒癌细胞系JEG-3,经不同浓度和不同时间TGF-β1作用后,采用Western blotting检测CD133表达情况,RT-PCR检测EMT相关标志物m RNA表达情况,并采用Transwell侵袭实验检测TGF-β1对绒癌细胞迁移能力的影响。结果 TGF-β1可促进绒癌细胞中肿瘤干性标记物CD133的表达,下调上皮细胞标志物E-cadherin m RNA的表达,上调间质细胞标志物N-cadherin m RNA的表达,并可增强绒癌细胞的迁移能力。结论 TGF-β1可增强绒癌细胞的干性特征,促进其上皮间质转化并提高其迁移能力。Objective To explore the role of TGF-β1 on epithelial mesenchymal transformation and invasion by promoting cancer stem cell marker CD133 expression in choriocarcinoma cells. Method Choriocarcinoma cells JEG-3 were cultured in vitro and incubated with TGF-β1 at different time and concentration, and the expression of CD133 protein and m RNA of EMT markers were detected by Western blotting and PCR respectively. The effect of TGF-β1 on invasive ability of JEG-3 cells were assessed with transwell method. Results TGF-β1 promoted the expression of cancer stem cell marker CD133, downregulated the epithelial marker E-cadherin, upregulated mesenchymal marker N-cadherin, and promoted invasion ability in choriocarcinoma cells. Conclusion TGF-β1 could promote stem cell property of cancer, EMT property, and invasive property of choriocarcinoma cells.
