摘 要:目的探讨体外培养神经元在缺氧时细胞红蛋白(cytoglobin,Cygb)基因表达的变化规律。方法运用无血清培养技术进行胎鼠皮质神经元培养6 d,用免疫组织化学方法进行纯度鉴定,随机分为4组,其中3个实验组分别置于0.5%~0.9% O2中8、16、24 h,对照组置于正常环境。用CCK8试剂盒测试细胞生存率,用Real-time PCR方法检测不同时点Cygb基因表达的情况。结果神经元纯度鉴定纯度〉99%;缺氧后神经元Cygb基因的表达随时间增长而递增。结论体外培养神经元缺氧时Cygb基因表达增强,提示Cygb可能在神经细胞缺氧时发挥重要作用。Objective To investigate the expression and changes of cytoglobin( Cygb) gene on hypoxic neurons in vitro. Methods The primary cortical neurons from newborn rats were cultured in a serum-free culture system.The neurons were identified by polyclonal antibody against neuron specific enolase on the 6th day and were randomly divided into 4 groups for assessing cell viability by CCK8 and for observing the gene expression of Cygb by Real-time PCR at 0 h,8 h,16 h,24 h after hypoxic injury was induced with the concentration of oxygen between 0. 5% and0. 9%. Results The purity of neurons was more than 99%; Expression of RNA Cygb was increased in a time-dependent manner,and there was significant difference in each group( P〈0. 05). Conclusion The expression of Cygb in neurons increases rapidly after hypoxic injury indicating that Cygb may have an important function in the process of protecting hypoxic neuron injury.
